The reason people are interested in RGD peptide is because it mimics the
extracellular matrix and binds to the integrins, I believe.
I actually don't really care what kind of peptide it is. However I am kind
of planning to do some pilot experiments by myself and I prefer to use
something that can be handled easily. Therefore, I would like to have
something that is sovlable and cytoplasmic (which will be easy for
expression and require minimal posttranslational modification). While the

听说罗经常对学生说发不了10以上的文章就免谈毕业。而关于endostain的受体文章有
多少是争议很大的并在世界一流杂志上发表后却不能被重复的。以前就说是integrin（
cell文章）。现在罗永章大谈nucleolin，而其后续文章全由其实验室出来，有多少可
信？
刚才看了看，endostain的PI很高（大于10）。这种情况上应该可以是非特意靠带电特
性就可以结合不少蛋白吧。 |

比方说，很多体外的实验认为这个系统对于细胞运动是关键因素，
但是很多体内实验证据指出integrin 其实对于细胞在体内运动影响非常小。

Formaldehyde as crosslinker. You need to find the lowest concentration of
formaldehyde in an initial test. I have been using 0.1% formaldehyde in PBS
to crosslink proteins on freshly harbested cells, room temperature for a few
minutes, and quench with 1.25M Glycine. Then lyse cells, sonicate and IP as

蛋白，和小鼠同源蛋白序列identity 在80%左右，为什么大鼠和仓鼠就会好一些呢？序
列同源性也不低啊~
integrin也有不少鼠单抗的，这是怎么做出来的呢？

in alpha-catenin. 其它domains稳定些，不过也只需要10-30 pN就能几秒钟内打开。
talin所有domain都能在<30 pN下几秒打开。在成熟的focal adhesion里，单根
integrin上的力> 40 pN. 我估计在cell-cell junction, 单根E-cadherin linkage上
受力也小不到哪里去。看起来力的因素是不能忽视的。动不动就改变binding affinity
成百上千倍。 |

比方说体内的血细胞在血管壁爬行以及在结缔组织里穿行的时候，没有人观察到类似
focal adhesion plaque 这种东西。血细胞在体内爬行的时候虽然需要用到integrin-
based adhesion 结构，但是并没有那种一个个独立点的"focal" adhesion.
我还听说在体外之所以能看到这个结构是因为把细胞养在硬的介质上造出来的artifact

发帖数: 911 | 30 其实我觉得如果把focal adhesion定义放宽点就没有什么特别大的debate了。Focal
adhesion的形貌和intracelluar actin network的3D组织有关，而actin network的形
成又和ECM的力学和几何形貌有关。比如在表面上，fibrinectin coated surface的大
小，形状都影响actin network形貌和细胞形状。从二维到三维, 拓扑上的改变更为剧
烈。所以看不见通常在二维成像上定义的focal adhesion斑块也属正常。
可能更重要的问题应该是调控二维表面focal adhesion形成和稳定性的蛋白们是否也调
控三维cell-ECM相互作用和cell migration. 在三维, 过去已经知道会形成
pseudopodia. 这个结构调控细胞migration, 而这个结构的形成又被focal adhesion
proteins调控。如果宽泛地把通常的二维focal adhesion和三维pseudopodia看作同一
套调控cell-

focal adhesion的形状的问题，而是这个投影在三维的网状的integrin-based
adhesion 和二维的点状的focal adhesion是否用的是同一套调控蛋白以及同一种调控
方式并没有搞明白。在这个情况下，研究二维的focal adhesion是否还具有生理意义就
会出现疑问。
