，反倒还要花钱去做CE，花钱去RENEW。而inactivate是免费的，以后要activate呢，
再交上费用就可以了。如果2-3年内没有工作身份，好似inactivate更加在经济上合算
些。大家能否讨论一下呢？
如果inactivate 达四年以上，如果将来重激活，要不要补上以前四年的CE 的credit?
还是只补交两年内的CE？（假如执照是两年一续的话） |

，要在ER里面被protease切掉latent group才能activate。现在我的实验要用到那个
precursor，不知在哪家公司可以买到这样的growth factor呢？
另外FGF里面好几个都不是通过ER分泌的，那怎样得到它的inactivated form并且用什
么酶可以让它activate呢？

然后switch to a stimulus and apply it to this neuron for 60s, 然后再switch
to LB buffer and apply It to this neuron for 60s, 发现这个Neuron 的calcium
response 一直呈下降的趋势, 并没有按设想的那样在apply LB 后回到baseline. 请教
两个问题：
1. 为什么这个neuron在applied stimulus 后一直呈下降趋势，即使在applied LB
buffer 之后？
2. 我能得出结论： this neuron is inactivated by this stimulus?

最近回去的人的情况，如云老师所说，绝大多数都有2cns，如果只有一篇cns，那基本
上还有>2的子刊。这个不仅仅局限于胖老师经常说的结构生物学，而是所有方向都基本
如此。
当然这些新回去的绝大部分都是千青（有些不是的是因为回去那一年千青还没开始）。
网页上的Title有的是教授，有的是研究员，估计当时还没有统一。但校内一律都是副
教授，也就是相当于这边的AP。我还特地打听了一下，国内的TT制度好像没有Tenured
Associate这一档，清华5年考核通过了就是正经的full professor。
有些老师看不上国内的科研，我就仅列出两位回国后的publication，咱就不说颜宁等
这种Early T的个例了。说两个没太有名气的，还没有评上T的，各位老师可以参考一下
，看看自己有没有别人这么productive。

是female mESC吗？是的话，先做两个实验：
1）H3K27me3 immunostaing：可能发现small dot in nucleus.
2)qRT-PCR测Xist的表达，或者用FISH。
证实的话，就说明出现X chormosome inactivation了。
几个问题：
3) mESC的培养条件？serum,还是2i/LIF?
1）knockdown的mESC是否仍保持未分化状态，还是开始分化了？如果未分化，那么很有
可能你发现了一个新的调节X reactivation/inactivation的因子，做深了，至少是
cell stem cell.如果细胞分化了，那么X chromosome上的genes表达下调不奇怪，有多
大新意取决于你的这个gene
2）看一下这个基因的功能，是否已有先关报道，与X chromosome reactivation有关。
是histone modifier, TF还是在X chromsome上的新lncRNA? |

inactivation不一致。
按理说，在mESC中，X chromosome是active的，即XaXa，Xist应该不表达才对。Xist表
达时，会把其中一条X chromosome“包裹”起来，这条chromosome进而失活。X
inactivation一般指发生在female mEpiSC与分化的细胞中.
你这个结果与理论相反，X chromosome基因下调时，Xist竟然也降低。Xist在对照和
knockdown细胞中的表达分别是多少(RPKM or CT value)？
另一个问题，由于你用的培养条件是serum+LIF,这会导致heterogeneity.建议你用2i/
LIF.
knockdown mESC分化明显吗？ colony morphology改变如何，还是依旧维持与WT一样的
形态？
如果你确定用的是female mESCs的话，建议先做H3K27me3 staining,看看X chromosome
是不是失活的。 |
