on. So I had spent a few hours to try to find what is in FBS that we used
for cell culture. But did not find a good answer. I know IGF and TGF. EGF?

到100ul的PBS/2%FBS里。
请问这样的话，收集在PBS/2%FBS里的细胞是直接放在冰上，还是赶紧在干冰上冻起来
？一般细胞在加Trizol之前大概要放个2个小时。

我当年在上海卖试剂的时候，最喜欢的就是这种大批量买fbs 的工业界客户，卖一次一个月基本就不用干活了。给的价格也都是单独的。
FBS 还有一个问题是不稳定，不同批次FBS 表达蛋白的得率不一样，这个对工业界来说是一个很大的问题。 |

午6点钟离开实验室之前配？因为你完全可以在第二天早上配media，那个时候你至少有
机会了解还有没有其他人比你更急需FBS的，必要时你可以先配250ml，省下一只给人。
如果lz是怕被别人抢先用掉，这种心态就更不对了。
除此之外，lz明明看到了负责分装FBS的人，而且FBS又剩的不多了，为什么不直接跟这
个人交流，要知道写在黑/白板上的目的不是为了表现professional，是为了交流，该
说的时候不说，你的小辫子就已经攥在人家手里了。
事实上，像楼主这种做法，实际上是一种接近规则极限的做法，很多中国人都会无意识
地这么做，不信你到中国人多的实验室试试，这是一种最让人觉得不快的做法，这种做
法让所有人都会很紧张，因为很多试剂常常会在不知不觉的时候处于用尽的边缘，用的
时候会有压力。所以说，碰到试剂快要用完的情况，应该主动提出来，让多数人都了解
目前的状况，是发现者的责任，而不是像lz这样，情况已经有些紧张的情况下，还只是
简单按照最基本的做法来。万一负责分装的人没看见你在板上写的字，情况会更糟糕。

removed from these numbers but not games against non-AQ FBS teams.
These numbers might look very different if you remove games against non-AQ
FBS teams. I think the schedules of the 1-loss SEC and ACC teams will look
a lot weaker if you remove games against non-AQ teams. Many SEC and ACC
teams schedule 3 or more non-AQ teams, which artificially inflates the
schedule strengths for all the teams in these conferences.

好像NBA都希望看到湖人，热火，雷霆，马刺终极对抗，但骑士，太阳，猛龙，山猫这
些球队也要存在，这样才是一个整体机制。而且商业媒体和联盟组织者还得让这些小球
队有定期发光的机会。其实大会议和P5委员会也是多余的，因为只要长角，邮迷，巴盖
，圣母，FSU，USC，鳄鱼，河马，巴马，LSU，田七，这些球队纯竞技纯商业利益最大
化招生，那基本其他110所FBS学校一点点机会都没有，渐渐这个机制就失去活力和商业
价值。不上不下的玉米，VT，鸭子，小威，牛狗，塔木，大U就会受到地区大校的压迫
。再往下排的其他几十所大联盟学校更是永无无翻身之日。结果就成了CFB整体商业价值
最后就绑在几所关键学校手里，观众数量和赛事价值就会越来越单薄。
我觉得CFB足球其实冠军不是目的，没有独善其身的结果。让这个机制平衡存在且创造源
源不断地共同价值，才是FBS这帮大小头头们更关心的事情。

FBS里面有一种叫做fibrin的蛋白质，thaw之后，就会聚集，产生蛋白质颗粒
（black dots），随着培养时间的延长，就会聚集越多。不同批次的FBS，
不同品质的FBS，里面含fibrin的量略有不同。
不要把这些蛋白质颗粒filter掉，细胞需要这些蛋白质，因为可能有生长因子。
实际上，细胞很喜欢吃这些蛋白质，你会发现很多细胞里有蛋白质颗粒。
如果颗粒实在太多太大了，可以非常低速地离心一下，去除掉过大的颗粒就行了。 |

从293 细胞adapted的悬浮细胞。 我从invitrogen公司买了这个细胞和其相应的培养基
后， 在普通的incubator （没有shaker）里面培养，第一次复苏完全没有成功。后来
又向 invitrogen要了一管， 复苏时在 293F culture media里面加了2%的 FBS， 复苏
后很好， 但可能是因为加入 FBS的原因，大部分细胞贴壁了。 随后的培养中去掉了
FBS， 活着的细胞还是继续贴壁，游离的细胞都是死细胞 或者 不是很健康。 而且可
能是因为没有 在orbital shaker里面培养的原因， 有很多细胞团。
问一下论坛里面的前辈， 你们培养293F细胞时有什么需要注意的事项么？ 非常感谢你

growth factors; also, your cells are generally synchronized if there is a
growth arrest with serum starvation. You can skip the starvation, and
usually you'll also see an increase in phospho-AKT, ERK1/2 etc.
You may try 0.2% FBS 0.1% FBS etc. instead of no FBS at all.
Check the literature about the concentrations used for your particular cell
lines. 10-100 nM is a good start. |

更多的dish里的培养液变混浊，重新解冻细胞(100mm dish)，加入DMEM+10%FBS培养两
天后再次发现同样的情况，是不是整个Incubator都污染了？现在还有29个35mm dish在
里面，发现其中有三个混浊化，但其它的还是清澈的，不知道到底是什么原因？一开始
想是不是FBS或DMEM污染,但是并不是全部的dish都混浊，遂排除了这个可能性，所加的
待检测试剂也不大可能，因为如果试剂污染，全部的dish都会变混浊呀，到底是什么问
题呢？希望大家能够帮忙找找可能的原因及提供如何解决的办法 |
