我们的OD推荐的是 Bausch+Lomb Boston Simplus Multi-Action Solution 和 One
Step Liquid Enzymatic Cleaner (protein remover)。
我们一直在附近的walmart买(没注意到其他地方卖)。但是最近好像他们不在上货了
。于是想看看有没有可替代的护理液。google没有发现有用的结果。
我们每天清洗时是先用盐水冲一遍,然后放入simplus solution,滴一滴enzymatic
cleaner。你们也是类似的吗? |

因为靠热运动组装第一个蛋白质是困难的,所以有观点认为第一个蛋白质是根据某个自
然产生的遗传基因密码(DNA/RNA)组装出来的。这便是热运动组装第一个遗传密码/基因
的理论。
在俺看来,这是先有蛋还是先有鸡的问题。蛋白质好比蛋,基因好比下蛋的鸡。如果自
然界糊出一个蛋很困难,那糊出一个下那个蛋的鸡就更困难。
事实上,每三个DNA碱基对决定一个氨基酸,加上基因的头码三个,尾码三个,对应一
个100个氨基酸长链的遗传密码(306个碱基对),其自然产生的困难程度和自然排列该
特定氨基酸长链的困难程度基本一样(遗传密码更难一点)。且遗传密码翻译为第一个

complex太大,sonication效果很不好。但实验室另一个postdoc认为enzymatic
shearing会把dna切得太小,会miss掉很多东西,他认为这个方法只适合chromatin
modification。
很纠结,不知道是否值得一试。 |

some big bulls made at Harvard/MIT 15 years ago. Half of the isozymes of
the same class do not have enzymatic activity and likely just serving as
binding domains. Even few years ago, some reviewers still want enzymatic
activity from these enzymes to be measured (talking about not keeping up
with the literature). |

对,这篇1997年的nature paper,列在第一位的yan shidu其实是唯一的通讯作者,列
在最后一位的本该是通讯作者的David Stern其实就是当时部门里的大牛。估计情况就
是David Stern是大老板,但是对这个project其实没啥贡献,然后是作为通讯作者的
Yan shidu是小老板,带领Fu Jin等一干人做出来的,才会有这样奇怪的authorship。
这样看来,Fu JIn实质上是真正干活的第一作者,凭这样一篇nature paper获得博士
也就没啥稀奇的了。Fu Jin是1999年拿的博士,1997年的paper正好对景。
凭什么方肘子要看学位证明人家就亮给这个傻逼看啊。他家菊花不是都拒绝出示学位证
明吗?人家Fu Jin教授把学位亮给学校看就好了,气死方教主及其教徒们!

然后在哥大的大老板手下做了小老板,
找了同是福建医学院89年毕业的Fu J做了她的tech./postdoc.
SD Yan的确做得不错,Fu J干活应该也不错,但哥大的博士学位应该是没有的。
连SD Yan都只是说自己是哥大postdoc,Fu J就敢说自己是哥大博士。
区别就在一个在美国,一个在中国。
另外一点就是SD Yan应该在福建科研界有些影响,所以Fu J的问题也可能会不了了之。

水便说说我能想到的技术层面吧。
chemical entity:
小分子药物,集中在癌症、感染、CNS、细菌/病毒等方面,还是有的可做,同时不断有
新的target出来,不断有nature prod出来,但受专利的影响(过期和剩余的chemical
space),大多都是在维持生计。在临床阶段对于试验病人的筛选,以后必将维持下去
。相应的试剂诊断盒业务,大公司为了利润和专利,倾向于自主研发或者收购这些诊断
盒,所以会有大的发展,类似invitrogen的公司。
大分子药物(theraputic toxin),应该在未来有大发展,基于专利和市场的考虑。只
是其价格、稳定性和进入人体的方式,不如小分子方便。至于personalisation,就是
根据病人自身的情况通过克隆和筛选得到最好的个人用药,我觉得需要等待技术上的突

之前主要是基于X衍射或者核磁共振的技术开发和应用,拍一些单分子的裸照。82,91
,02年颁给X衍射和核磁共振的技术开发,剩下11个的给了应用。
而冷冻电镜,这次算是第一个(可能是第二个),直接给了技术。用它可以看更加复杂
、分子量更大的大分子复合体。这些大个头的结构当然更有复杂也更有意思。
这个技术开启了一个新时代,细胞里重要的复合体实在太多。接下来可能拿奖的那就是
:真核细胞的细胞核通道复合体;一一公搞的剪接体(这个结构无论在生命进化和疾病
相关都非常的重要);G蛋白偶联受体(跟N多疾病有关)等等。
彦宁的工作好像确实没有什么希望,葡萄糖转运蛋白跟上面这些不是一个量级的。
