San Juan Mt.位于科州的西南角，据称是科州最人迹罕至但风景最优美的山脉。前年夏
天开车去过一次，从此念念不忘。 说服了爬山伙伴小A，我们决定要into the wild。
本来没打算带小朋友， 无奈找不到合适的babysitter，再加上小朋友死乞白赖，苦苦
哀求。我心想不过是多走一天，或者少爬一个14er，就当路上多带个玩具解闷儿吧。
于是，我们两个半人，小A,我，和我家7岁话痨小朋友开始了六天五夜的探险。路程从
Elk Park顺时针到Needleton，总共大致50迈，两上两下，海拔从8700ft->12500ft-

当快到一个小池塘的时候，我这不速之客，终于被盯上了。一头年轻的驼鹿竟然就沿着
小路低头向我跑过来。
直到后来在小池边休息时，我看着它在那儿开心的啃着青草，又看了看自己手上的肉干
，还是没搞明白到底谁是肉食性动物。
第二天，也是一个晴天。继续走着上坡路。大的动物没怎么见到，小的却有一些。
渐渐的，顺着山花和小溪，心慢慢的随着周围的声音安静下来。

车子终于驶上了开往teton的公路，离开黄石，我们是那么地恋恋不舍。在快出公园南
门的地方的路东边有一个牌子，上写着 Continental Divide. 公园内的公路大多沿河
而开，可是有时候明明刚沿着一条由西向东流的河开，没多远，沿着公路的河就变成了
由东往西流，开始我怀疑自己看错了，河水怎么会倒行逆施呢? 这条continental
divide就是这个疑问的答案。原来由于北美大陆的continental divide南北纵贯黄石公
园，在黄石内流经的河有些向西注入太平洋，比如madison river, snake river, 有的
向东经密西西比河注入大西洋，比如yellowstone river。
开往teton的路大多是沿着snake river的。teton中最大的湖是jackson lake. 由于我
们今天有比较长的路要赶，在teton没有作过多停留。在jackson lake的一个观景点，

发帖数: 3745 | 25 两周前为了装tank divider偶然发现鱼缸顶部的塑料大梁断了，没有了大梁的拉力鱼缸
的长边在水的压力下开始向外鼓，本来直的玻璃向外弯成了一个大弧线，在网上一查说
随时有可能爆缸。55加仑的鱼缸爆缸那就几乎是个小规模发水啊！
危机当前，家里的危机处理委员会小组（只有小组长光杆司令一名）立刻开始工作。用
变形鱼缸的水和部分底砂建好了一个闲置的10加仑小鱼缸，幸好之前已经配好了加热器
，过滤器，空气石和气泵，并且事实验证了均工作正常，使得小鱼缸立刻就成功启动。
把小白和小清道夫转移到小鱼缸里后，立刻把变形鱼缸里的水全部泄掉，哇，松了一口
气。
之后的工作就不是那么着急了，危机处理委员会小组长摇身一变为危机后续清理重建委
员会小组长，按部就班的开始清理废旧大鱼缸的工作，包括卸除所有底砂（装满了两个
homedepot 5 gallon bucket），拆卸清洗过滤器，把鱼缸从底座上搬下来转移到车库
冲洗之后闲置。现在还没来得及买新缸，于是就把木头顶直接摞在木头底座上了，如图
1。
而我的工作，就是照顾新建的10加仑小鱼缸。除了确认水温刚好在78华氏左右，最重要

具体说说我的virus载体和transduction的过程吧~
1> retrovirus用的是MSCV based带有mir155 backbone的一个载体（某postdoc自己构
建的）有GFP marker。因为它只侵染dividing cell, 所以就在collect BM的当天做一
次spin infection （用fresh virus containing sup from transfected 293T cells)
，之后两天各做一次spin infection，然后让细胞在有GMCSF的medium里面继续分化到
day7 (中间换一次medium). 最后收细胞染色FACS. 现在结果就是no virus control分
化没什么问题，通常我得到90％ CD11c+细胞，其中MHCII+大概15％；问题是无论是
control virus (empty vector)还是有target shRNA的virus，细胞就不表达CD11c了 (

公理：Idea都是testable 的。否则叫幻想，宗教，或白日梦。
好idea 第一定律，如果成了，收益远远大于投入；第二定律，只有少数人能想到。第
三定律，需要test时间很长的idea，有divide and conquer的可能。
举例：第一点，如果某个idea要花几个月test，成了就是一片好文章或grant的基础，
那就是好idea。如果要花五年以上test，要成了能得炸药奖或Lasker 奖那才值。好
idea如果成了能开辟新领域或至少现有领域能更深入，而不是平行地挖类似的坑。第一
点，在试成之前，没经验的人很难判断好坏，所以就有了第二条。
第二点，如果一个idea 只有你想到了，实验室其他人都没想到，那就可能是好idea。
这个要求你有平均水平以上智商。智商太低的，基本上没有别人想不到而你想到的。
第三点，不一定总成立，有的好idea不能分割成更小的部分，但大多数在生物学领域的
好idea都是可以divide and conquer 并且能step-by-step logically test。也就是说

lenti packging can be used for Retro(pbabe). So for dividing cells, doesn't
matter much for the infection/integration; However, for non-dividing cells,
presumably no big difference with infection but a huge difference with
integration step(nuclear entry step--which is essentially the key difference
between the two)...So essentially western doesn't tell you much here...
unless they stablize.

1）要做一个实时的数据通路，input有24位，output只有16位。不考虑用先编码后解码
的方法。其中要过一个ram，想到过用32 input/16output的ram，并设置不同的in/out
clk频率。那么是不是说input的clock就应该是output的clock的一半，才能保证数据的
实时传输？用ise自动生成这样的ram后，读写controller应该就分开写吧？
2）采样一个信号，比如用简单的一个2级串联的FF来采样（没有试过，不知可行否），
要求采样频率可调。我用不同divider生成了不同频率的clk，再用mux来选择这些clk，
会出问题否？因为听人说过不宜用mux来选择时钟信号。另外，如果我只是用behavior
来写一个可调的divider，实际生成的电路是不是也是用mux来选不同divider生成的不
同的clk？
3）现在要求采样长度可调。如果用上述FF的方法来做，怎样做到长度可调？比如设置
采样100个clk，采样50个clk，采样200个clk。。。。

我觉得你将两个概念搞混了， 一个是无穷集合的势（cardinality）， 一个是集合的
测度(measure)。
one to one and onto map 和集合的势密切相关， 但是和集合的测度几乎没有关系。
一点典型的例子就是R包含不可测集， 显然这个集合是和R等势的。
问题是什么是测度呢， 基本上就是测量长度， 体积等等的推广。 所以测度差不多意
味着

% 的人是携带者。做 Rapid Strep 或Culture 会阳性。
这里有一个问题，一般三岁以下的孩子，得strep Throat 的比较少。通常是携带。
现在我们多Strep 阳性 实际上是 过度使用抗菌素。但也是情理中。
他的药物问题是这样的：如果使用Amoxil治疗Strep Throat ， 没有 QD 的 使
用建议。是 BID 或 TID 。使用剂量是 ２５－５０ ｍｇ／ｋｇ／２４ｈ． 分
两次 或三次。他是 ３６磅。合１４ 公斤。 大约日剂量是 ７００－８００ MG
。 这样他要两个ｔｓｐ （５ｍｌ／ｔｓｐ）每天。分两次的话，要一次一个Tsp.如
