发帖数: 94 | 5 很多long ncRNAs本身就比较短，表达量又低不容易检测。所以担心150bp paired end
建库的时候就是300bp左右了，容易把一些lncRNA给弄断了弄丢了。所以我的一个很个
人的建议是，直接100-nt SE就可以。重点是测得深一些，后续分析仔细点。
至于exon-exon junction，其实也无所谓了。 long ncRNA gene的本身是热点，但的
splice variant其实感觉上也不是一个特别好的热点。因为做这个的需要后续，这时
候麻烦事或者无效劳动量也挺多，个人意见了。
还有就是strand specific library之后用Cufflinks这一套去拼接lncRNA。这个如果是
intergenic的，那还好说。如果是Antisense，这个如上文有人提到，有"很多tricky的
地方"，从tophat这一步就开始了。
另外就是Poly-A selection或者Ribosomal RNA depletion。其实这两个技术回答的问题
有点不一样。 如果没有什么特异的考虑，那么我个人觉得Poly-A selection鉴定

induce tumor。当然，大部分tumor里面p53是loss of function的，所以想做药肯定不
是用inhibitor，而是想办法恢复功能，但是小分子很难有这种功效的。
Cancer stem cell是一个很异源性的population，不同cancer里CSC的起源可能完全不
同，甚至在同一个体的tumor中都有可能存在不同来源的CSC。有些cancer，比如某些脑
瘤，deplete了CSC以后，不会再形成新的CSC，而有些tumor，比如breast cancer，
deplete了CSC以后还会形成新的CSC，甚至CSC会从triple positive 向triple
negative发展。CSC的复杂性很大程度上也是和cancer的多源性有关系的。
个性化治疗对某些发病机理相对简单的disease还是很promising的，而且有针对的治疗
至少可以improve survival和life quality。 |

One version I knew refers water flood as IOR - seconary depletion, and then
gas injection etc. as EOR - tertiary depletion.
When people envy the current high salary package for PE's, please don't
forget the massive layoffs in 1990's when crude oil was below $20/bbl. And
PE's have very limited job opportunities outside energy industry. So it's

看paper里测CV的时候，最后分析电路基本都是按照附件里的电路图来分析的，有几个
部分，C(I)代表insulator的contribution,C(D) is the contribution from
depletion region between insulator and semiconductor, C(B) is for the bulk
semiconductor, R(C)is for the contact between semiconductor and metal
electrode

1.为什么R(B)和C(B)画出来是并联的而不是串联的呢？

3.还有一个比较相关的问题---我看很多device里面insulator可能挺薄的，只有几个纳

论多么奇怪，什么大乘小乘，唯识唯物，秘传显宗，甚至连轮回转世都可以否定不要（
比如汉传禅宗），都可以戴一个佛教的帽子。
这几条里面，有的是哲学世界观（诸行无常，诸法无我），有的是佛教提供的救赎道路
（涅槃）。但是有一个共同点，那就是和科学无关，也就是和科学认识论是平行的，不
能证伪，当然也不能证实。所以科学永远打倒不了它。
如果科学能打倒，其核心理论能被证伪，那就是不是宗教，而是迷信了。这是我认为的
：宗教和迷信的一个本质区别。
其它所有宗教，都有一些核心论述。同样，也都有一个庞大的，几乎可以无限外延和发
展的体系。只要想象基督教的浩如烟海的神学，佛教的堆积成山的佛经和注释，伊斯兰
的“法学”，道藏丹经，印度教的，，，等等。

I find the sanctimony and hypocrisy coming out of Washington very annoying.
"Chemical weapons!" which of course are nasty, cruel, and inhumane, have
been framed as the ultimate affront to all morality.
But land mines, which the US refuses to eschew, and cluster bombs, which the
US also refuses to eschew, and so-called "collateral damage" from drone
strikes -- meaning civilian maiming and deaths of old people, noncombatants,
women, and children -- somehow escape this calculus.

distances in perspective. The scale bar is measured in astronomical units (
AU), with each set distance beyond 1 AU representing 10 times the previous
distance. Each AU is equal to the distance from the sun to the Earth. It
took from 1977 to 2013 for Voyager 1 to reach the edge of interstellar space.
