因为有些效果值超大或者超小，我怀疑是样品的浓度有点问题，所以我想把浓度和效果
值之间做一个paired Wilconxon test

we order from Invitrogen, and they use different salt concentration
to calculate Tm. It's not about right or wrong, coz Tm changes with
oligonucleotide and salt concentrations, so u'd better calculate it by
yourself. Some softwares like Vector NTI can do it, I think Primer3
works too.
we use oligo conc. at 250 pM and salt conc. at 250 pM to calculate
the Tms. |

0/2/5/20/50/100 ug/ml 的target protein，RNA MW ~30kD，protein MW ~200kD，PI
~5.3。用的是6% PAGE gel，没调pH。
主要有两大问题：1. free RNA band（bottom bands）是smear，而且band intensity
非常
低。不知是不是因为gel conc.太低的缘故。
2. 如果只是这一点也就罢了，从RNA-protein complex band应该就能得到kD，可是
0protein
smaple就有两条band，杯具。。。
之后做了点troubleshooting: 1. run了其他两个RNA sample，也都有两条band，所以
不是某一
个sample screw up；2. 怀疑protein-RNA complex没有入胶，改用4%gel（以前做过
staining，free protein 4％ gel之下肯定能入胶），结果和附件基本一样。

before u propose protein-ion interaction, I think u should make sure the
precipitation is ur protein and it is phosphate-dependent.
use HEPES as buffer and add series conc. of Pi to test dose-response. Only
difficulty is to quantify protein precipitation, but u could spin down it
and determine protein conc in supernatant. |

一错到底，没有回旋余地。我刚过去，再说他马上要走的人，也就顺着他来。直到他走
人了我再和老板提高这个致命的实验缺陷，做了改进。
后来其他同事和导师的评价基本验证了我前面的说法。两个典型的例子。有一个同事请
教了他一个问题，他后来就利用这点，逼着同事给他的样本做实验，具体操作就是威逼
利诱。另外一个，他指导一个学生做实验，学生做了实验，写好了文章，结果他要争第

Cracking was noted at the right side foundation wall (see attached photo).
It appears to have been caused by expansion and contraction and would be
considered typical for a poured concrete foundation.
看起来不是个大问题，可是还是有点担心，毕竟是在foundation wall上，裂缝会不会
变大，值得让seller来fix吗？这是个大问题吗？如果自己fix的话，大概多少钱。
