境的融合。真正赏心悦目的花园不一定要非常恢弘、工整，而是适度的、成熟的，并且
和房屋、环境融洽的。
我所居住的城市在1900-1950年代曾经辉煌，我曾经居住过的这条街道上，都是一些保
存完好的1920's Colonials，每家每户都有精心耕耘的花园。从大路一转进入这条静谧
街道，每家每户的房屋和花园都有非常浓厚的architechture detail和历史积淀的感觉
，田园气息浓郁。

PCR总是令人烦恼，国内的时候就遇到过，现在又出来折腾我了：
我是想分别扩e.coli两个蛋白的两个同源domain,都很小，200bp的样子，但总是得不到
，都只有引物二聚体。我不觉得是引物的问题，因为两组引物都失败了，而且以前没有
遇到过这个问题。我到觉得是模板的问题。我直接用的colony，然后用PCR的那个95度
10Min，理论上认为能裂菌。我在想是裂菌不充分呢？还是我开始加的菌落太多了？我
一直担心模板不够，挑个菌落在tube里twirl了很久。或者裂菌之后就是问题比较多，
我应该尝试的先制备chromosome再扩增呢？太头大了，牛人过来指点下吧 |

1. 挑一个colony加到5 ul solution 2 （plasmids prep）；
2. pipette几次确保细胞裂解了(可以加热到100C for 2 min)；
3. 加入0.5 ml 10 mM Tris-HCl (pH 8.0) or H2O;
4. 15,000xg for 5 min;
5. 用上清作为模板(0.5 or 1 ul)；
6. 这个上清可以保存在-20C供下次再用。 |

爬这几个山头是年初就和山友T计划好的，因为T的工作性质，请假必须提前好几个月通
知，我们就把日子定在奥运开幕前的那个周末。至于到时候天气如何，只能撞大运了，
反正爬山本来就是靠天吃饭，三天之后的气象预报就不怎么靠谱，索性来个拣日不如撞
日。行程如下：

发帖数: 349 | 25 新手，最近在做一个质粒的稳定转染至肝癌细胞，大概步骤如下，6孔板中转染，24小
时后按1:50的比例传至10cm dish中，（大约1~2万个细胞），再等24小时开始加药筛选
，没隔3天换液维持，至第9天前后开始出现大量细胞死亡，并出现镜下明显可见的
colony，至第12天左右出现肉眼下明显可见的colony，于是用黄色tips直接在肉眼下挑
取单个colony，分别转移至96孔板中培养，并逐渐放大至24孔板，然后进行鉴定。至今
挑了大概40个colony，仅有一个阳性，但是继续放大培养两代后阳性反应突然丢失。
现在我有两个问题,
1）阳性反应突然丢失的原因可能是什么？细胞不纯？阳性细胞被其他细胞取代？筛选
出colony并放大培养的最初几代过程中，是否一定不能中断选择药物？哪怕只有两三天
的中断？
2）在网上翻了些帖子，发现有的人会在挑出单个colony培养贴壁后在做有限稀释，挑
取单克隆，这样做是不是会是拿到真阳性克隆的机会更大？可不可以按照这样的思路来
做，先用我上面的挑取，培养并鉴定出一批（视运气好坏多少不等）有阳性反应的

Britain did not levy any tax on Hong Kong. It worked the other way round.

我回去也查了资料，我俩有一个共同的错误，那就是对殖民地时期的dgp盲从。有本书
叫做：Slavery and American Economic Development，这里面提到per capita GDP is
to some degree unhistorical in that it rrepresents the modern egalitarian
perspective. 书中后面解释了为什么dpg不适合来解释美国建国初国家经济。一个是奴
隶没有投票权，可是却要计算在dpg per capita里面。二个是gdp是当代的流动性概念
（你说你有一块钱产值，那必须我这边也认你是一块钱产值，也就是你的商品在我这里
也值一块钱），而建国时的国家经济很多是没什么流动性的（区域的价格都不同，实际
