of RNA ionize with H+, and is soluble in aqueous phase. While DNA is double
stranded, it's base is not exposed, it's not soluble in either aqueous or

不走column我就怕有污染。。。
我有时候担心吸到aqueous phase下面的白白的东西，所以就宁愿浪费一点aqueous
phase,但是如果换个小号的tips，就会吸得干净些，产量也会高些。

这个星期第一次做CHIP.完整的protocol就不贴了，但：
1. reverse crosslink 在65℃上热了16个小时。
2. 然后加proteinaseK在37℃热了2个小时。
3. 纯化DNA用的是phenol/chlorofoam.做这一步的时候有点怪。加了phenol/
chlorofoam然后spin down，可以看到aqueous layer和organic layer,但中间的那层
lipid layer基本上就没有。
最奇怪的是phenol/chlorofoam的最后几步。我把aqueous layer提出后加ethanol+NaCl
+glycogen, 然后放零下80℃三个小时后spin down, 居然看到一个有我半个小拇指指甲
大的白色的precipitate.当时我就觉得有点不对劲。但我用70%ethanol洗了一遍之后，
这个precipitate的体积马上减少了90%多，颜色也变蓝了（我们用的glycogen是蓝色的
glycoblue).之后我再洗了一遍。
最后我用nanodrop测，发现所有sample的260/280都只有1.... 阅读全帖 |

The source apportionment is done by chemical composition analysis, not the
proportion of aqueous and solid phases......
You really don't know what you are talking about. There is no real-time PM
monitor that can tell you the increases in two phases. The only way you can
quantify/differentiate them is to look at the collected PM samples using a
scanning electron microscope.
