我也觉得computation modeling应该大力发展作为最有效省钱的方式,但是不清楚到底
现阶段modeling and simulation有多靠,比如听说过一点use of bayesian model to
predict novel protein-protein interactions. 但是似乎这些modeling更多是学校的
理论阶段,拿一些数据train一下model,但这些machine learning 推广到未知有多少的
预见性?

尽量收集病人的数据 最好能把基因数据都分析了
现在肯定还是做不到 但是我觉得应该向这个趋势发展 |

,这个也是一种modeling吧。我前面帖子的意思更可能倾向于怎么利用in-silico
simulation来筛选分子,大分子,小分子,大小分子结合都算上。
有没有人知道现实工业界中这种成功率多大阿?还有大家这个依赖的程度多大?向楼上
某位说的基本上是给一个概率方向,那decision making得时候权重多少呢?sorry, 这个问题

验数据。底层是药物靶点作用,上层有pk/pd, resistance, 靶点的表达量,还有病人
的个体差异。每层模型都有偏差,层层叠加后可信度应该会大大降低吧。

up recently??? Just a few ADC drugs (should be less than 10) are in market
from then to present. If the advantage of ADC is really what it is supposed
to be (highly specific/efficacious, prolonged half life, less tox issue/side
effect etc.), then what's the hurdle that prevents the development?
Insufficient understanding of biological mechanism of action is the same
problem for developing any type of drug, so the long cooling time must be
from something else, i.e in-vivo linker stability, immunogenicity??
Really appreciate if someone could offer an opinion in this regards.

Often times the linking is random on the mAb surface
and the linker is not very stable
so in vivo the drug is dissociated from the mAb before reaching the target
But there is some progress right now, Genentech invented a way to link drug
to specific peptide residue.

Right, Cys conjugation is one of the major types, either through partial
reduction or using an engineered Cys like you mentioned. If we really want
to pick some flaws, is there anything bad from the engineering? like
inconsistent scale-up? Also, how does the thiol group linked to melamide
behave in vivo? I once went to a siminar by someone from genentech talking about pk/pd, looks like there is still some amount of free drugs in serum.

purification, or reversable linkage chemistry. There are always trace free
thiol. |

total Ab assay and the assay to detect the drug. And then they actually
showed how to use LC-MS to measure the residual toxins.
Well, maybe they just intended to use a bad example so they could give a
public talk:-)

模。现在基本每个公司都在做ADC.
Wyeth的mylotag在被pfizer withdraw以后现在又准备refile,说是新的clinical数据非
常好。还有几个二三期临床的ADC据说效果很好。

作。问题是多方面的。以前antibody的技术就不成熟,用的toxin的毒性也不够强,
linker的性能也不够好。一直到最近几年技术成熟了所以才又热了起来。
