已经有shRNA转入细胞，抗生素筛选已成。需要doxycycline induction去减少一个蛋白。
没用过doxycycline induction。
1。doxycycline都用那个公司产品?Cat#是什么？用什么试剂配母液？
2。doxycycline induces多长时间（1天？2天？。。。）才能有反应，从而进行后续试
验？
3。doxycycline induction是在regular medium里进行，还是serum free medium里进
行。

来我订对了！
他们的液体也是在DMSO里溶的。
既然你们已经可以用水溶，我也用水溶，这样对细胞伤害小些。
水溶的信息很重要。太感谢了！
另外，母液存放的多少度？-20C还是4C？ 谢谢！ |

那么再用药物处理细胞时，是否还要让DOX和处理的药物共存在培养基中？
DOX如果处理一周，细胞会死亡么？
针对你们的细胞，DOX的浓度是多少？

concentration to see our protein expression level. Higher dox does induce
more our protein expression. So if you prefer a certainly level, you may
have to test.

Never tried that long. 2 days with 2 ug/ml would not kill 293 cells at my
hands. Dox has low toxicity to mammalian cells. I think it might be ok.
However, I would suggest you to test dox alone.
1-5 ug/ml is recommended. I myself used 2 ug/ml for tet-off system. For tet-
on system, the protein expression level might related to your dox amount. so
you better test. |

24h DOX Induction, then 3 days Drug treatment with DOX.
至于蛋白量需要WB 检测。但是MTS显示了我预期的结果。显微镜下也看到了细胞形状的
变化(Apoptosis/cell death)。
Negative control and shRNA knockdown 在细胞viability上有着显著的差异！
