phosphorylated substrate motif, termed the Cdc4 phosphodegron (CPD）。
但它本身受不受磷酸化调控我不是很清楚。 |

Ub E3 ligase的数量很多很多，那时已经知道有500个之多了，可以媲美phospho
kinase 800个之多那个数量，两者均为繁复调控的手段
E1 大概12个, E2 50-80个
所以，这么多E3，可以想象是很复杂的故事 |

To achieve high substrate specificity, cells express many different E2
enzymes (about 30)
and even more E3 ligases (more than 300).
The latter are categorized into four major
classes according to the presence of particular structural motifs: HECT-,
RING-finger,
U-box and PHD-finger-type E3 ligases (reviewed in Nakayama and Nakayama 2006
).
RING-finger-type E3 ligases comprise the largest group and are further
subdivided into
subfamilies. Among these, cullin-based E3 ligases are the largest single
class of E3s.

发帖数: 2041 | 11 What's the best assay to test a enzyme's ability to ubiquitinate. I'm
studying a well know enzyme with two catalytic sites and I found a meeting
abstract saying one of the domain has nothing to do with its normal function
but it is a E3 ligase. The full paper is not published, so I don't know
how solid the data is. I have the recombinant enzyme plus mutants for each
catalytic site. The interesting part is the enzyme itself have been
published to bind several known E3 ligase, I wounder the abstract author has
rule that out or not. Both the PI and the post-doc are Chinese so I think
they read this board, want to collaborate? I'm more interested in its
inhibitors. |

以human为例，目前只知道两个E1吧？E2 40-50个，E3 500-1000个。

准确。
