You need to learn how to turn people down.
First of all,you should never let him know that you have all he wants in the
first place.
Pretend that you don't have any left.
For antibodies, make aliquots and store in your own place.
If he asks for it, tell him that you are on your last vial and just keep re-
using a diluted tube. Tell him that you can share with him the re-used tube
but he has to return it so that you can continue to use it.
Nothing he can complain about.

所以几个星期前从ATCC买了，发现运到的frozen vial 已经被一种真菌感染。
现在在跟他们交涉。请问有没有实验室从其他地方买这个细胞株，长势健康的？
我们实验室一直从ATCC订细胞，从来都不错，不知道这次倒了什么霉运了。
如果其他地方／公司口碑比较好的，希望推荐。

我的方法给你参考。
E13.5或E14.5，去头去内脏，转到一个预先装有0.25%Trypsin-0.53mM EDTA的3mL注射
器里，从20G的针头中挤过。如果组织块太大的话，多挤几次。然后在37C消化5分钟。
用FBS终止酶消化，离心收集组织和细胞，用5mL pipette重悬。每1-2个胚胎可以铺一
个10cm的dish。一两天后就长满了，这算P0。需要注意的是所有和组织接触的塑料器皿
用前需要先润湿，否则组织很容易粘到上面。
传代用0.25%Trypsin-0.53mMEDTA，37C，3-5分钟。这算P1。从这里起，seeding
density在4-5 X 10^3/cm2. 3-4天或者接近长满的时候分一次。一般在第2或3次的时候
冻足够多的细胞，1x10^6/vial. 运气好的话，一只孕鼠的6-8个胚胎分出来的MEF足够
你用的了。 |

There might be money in it so some lawyer might be interested.
He shouldn't even plea for illegal access of the server. If there were no
official notification that bars him from accessing the server or deleting

requirement), then there is nothing illegal in what he did.

，因为我知道这样T细胞不会有事。但同组的同事很反对，说lab过去的cancer cell都
是用DMEM，我如果改成RPMI，就很难跟其他data比较。所以很头大。 |

如果你用的是LC-vial,可以放个100-200uL的insert在里面，如果样品被吸走了，液面
高低变化会很明显的。
如果你用的是96-plate,可以选择V-shape 1 mL或者2 mL，只放100 uL样品进去，如果
样品被吸走的话，从Plate底下往上看，液面变化也会很明显的。
你说的用天平称这种事，我也干过。以前公司里没有plate reader，我calibrate
liquid handler倒也这么做过 |

不能挥发掉，什么样子的瓶子最合适？普通的vial加上四氟乙烯胶带甲醇一天就没了。

2. 老B跑过来跟我说，你要做三个反应，你看Will，每天做十个反应。Will的Benc
h跟我不在一个实验室，我当时只觉得自己为什么这么不努力，为什么做得这么慢，
因为我每天在实验室，一般也只能开到两个左右，只有一天到了三个。直到我离开
这个组之后，才知道，所说的那十个反应都是在vial里做的，都是trial reaction

你们有人能做到三个，我做不到，我笨，所以我不学化学了。

我不知道大家的实验纪录是怎么写的，普遍的要求怎么样。丫给我看的范文，我每

今天最后一天。
我在NMR谱图上贴标签的时候，手写分子式，比任何时候都要认真。也许我以后一辈子
也不会在写天然产物的分子式了，呵呵。11个手性中心，3个顺反双键，两个环，还有
一个moiety 怕酸，另一个moiety怕碱。不算复杂，但也不好做。看着我的几大本子谱
图，几大本子实验记录，冰箱里一盒一盒的样品，无论以后他们怕酸，怕碱，怕光，怕
水，怕氧，不能久放，再也不关我的事了。
这么些年，我纯手工人肉过得柱子有两千根了吧。。。几年下来，发了三篇一区一作的
文章（mitbbs的热心读者请勿评论），有全合成，有方法学，还有全合成加方法学。写
到这里，真的眼泪要出来了。不为这些今后再也无人问津的所谓“科研成果”，只为我
的曾经的t-butyl lithium的小火花（mitbbs的热心读者请勿评论），大瓶大瓶的HMPA
的蒸馏纯化，必须每次新鲜制备的硫醇，大量的Raney-Ni, azide, 和每每抬头从小窗
里望到的满天星斗。你懂的。
