背景:
在一个生物技术startup (<50人) 的一个核心组作一个scientist.
公司整体的业务是,我们组设计产品, 加工组把它画成图纸然后委托外部加工一定批量
然后交给我们测试. 我们测试之后再改进. 在测试过程出现问题可能要借助其它检测手
段和仪器才能解决. 具体的手段要随时看新的科研进展.
公司科研副总和工程副总两人负责管理. 两人都年近六旬,科研能力不能保证完全统领
全公司.因此在不同人在技术原理和思路上有不同主张的时候,不作出谁对谁错的仲裁.
领导关系:工程副总->我经理(Ａ)->我和另外两个同级别同事(包括同事Ｂ). 科研副总-
>其它组->其它人(包括Ｃ).
我(男): phd毕业在本公司工作两年,无卡. 公司唯一一个某核心技术科班出身. 刚来做
测试苦力. 慢慢吃透整个组设计的原理.在很多软件,理论上掌握领先地位, 独自代表公
司外出开学术会议. 入职两年半.

1。先去买个ODB/USB连接线，含运费不超过20刀, http://amzn.to/zf9lOY 。
2。到货后把连接线的USB插入笔记本电脑（这时不用连汽车），电脑会提示有新硬件需
要驱动程序，这时把连接线随附的CD装上，电脑会自动找到所需的驱动程序。然后去电
脑countrol panel/device manager/Ports, 把USB Serial Port 设成5，这样不会和已
有的port冲突。

网和电视服务连在一起，听上去好像比Comcast划算很多，月费不到$40,很动心想从

怀疑仙5没有1和4好。其实4也没有1好，1里面的林月如挺好的，形象简单经典好看，赵
灵儿比较一般，因为她基本上就是个touble人物，甚至阿奴都好一点。
4里面的菱纱和梦漓人物形象有点奇怪，感觉也不太好有点怪~~不过慕容紫英还不错，
比李逍遥好很多。云天河很一般。

昨天做了一个类似的，只不过我是5k + 5k
I assume 你gel extraction是干净彻底的
我用的molar ratio 是1:1，我不知道你为什么要用3:1或者其他的。为什么一定要认定
一个是insert，而另一个是plasmid呢，为什么不能换一下？
我用的是NEB的普通T4 DNA ligase， 连接1小时（普通的我一般是连接20min，也值按
照Protocol上说的），因为片段比较大。
I assume 你的感受态是没有问题的。
最后我提醒一下你，有时候酶切验证会失败，原因是不你克隆失败，而是其他原因，比
如甲基化。
Touble-shooting一下，不要漫天考虑太多的因素。
