比如美国著名的SR-71黑鸟高超音速飞机因为飞行时空气摩擦的高温把机体设计成亚音
速时是漏油的，为了发动机的安全而使用了高闪点特殊燃料JP-7，这种燃料非常难以点
燃，所以发动机启动时需要喷入遇空气即自燃的TEB作为启动剂。SR-71的多工况发动机
每次改变工作状态（涡喷-冲压）都需要喷入TEB启动，而其携带的TEB是有限的，所以
SR-71的每次任务都需要非常详细地事先规划好什么时候进行何种模式的飞行才行。 |

1. 要配15%的胶，注意组蛋白分子量10多K，不要跑出去了。
2. 酸抽：收集细胞，PBS洗干净；用TEB buffer（TritonX100 extraction buffer：
PBS＋0.5% TritonX100（v/v）＋2mM PMSF），冰上裂解10分钟，每10X10^6个细胞用
1ml TEB buffer。6500 g，4度离心10分钟，去掉上清，pellet就是nuclei。加0.2N
HCl（in水溶液），重悬起来，转轮上4度过夜，每10X10^6个细胞用250ul 0.2N HCl。
第二天6500 g4度离心10分钟，保留上清，就是histone。盐酸中的组蛋白，保存在－20
度、biorad测蛋白浓度，都没问题，不用TCA沉淀。
3. 一般取2-5ul，直接加SDS loading buffer就可以跑胶。不用改pH值。加了loading
buffer溴酚蓝会变色，因为pH太低，不过无所谓。跑胶转膜，ponceau S染色就能看到
