synergism 就越好。手头有一两百种药的某几个癌症的 cell line 上的 microarray
数据以及 IC50，那么就要先用图论的工具和几种备选的图模型来两两计算药的相干性
，得出基准分布，进一步算出每对药的组合的大家深恶痛绝的 p-values，然后根据要
使用的统计模型设计抽样方式来保证正交性的同时尽量减少样本量。然后上 384 孔板
，测数据，用事先选的统计模型分析来看哪种图模型最好地符合 hypothesis，同时看
这时 hypothesis 是否靠谱。然后用这种图模型预测最佳和最差的药的组合，重新上
384 板验证。在图上找到最佳组合的关键节点，k/d 或者 inhibit 之，再上 384 板验
证。最后根据经费和信心，要么 in vitro 上病人的细胞，要么 in vivo 在老鼠上种

curves是比较现代的用来发表的东西，你为什么要转化？
就这个图来说，B很可能是negative allosteric modulator，因为competition curve
达到的plateau跟你用的agonist有关，说明B不是competitive binding。如果B是
competitive binding，理论上当B达到足够的浓度应该会完全compete掉agonists从而
最后的plateau完全是B的效果，如同A一样，agonist-independent，现在B的
competition curves plateau是agonist-dependent，说明B是non-competitive
binding。
当然，证明一个compound是allosteric ligand是复杂的，一个实验不够，往往需要多
个实验去从各个方面证明。就你贴的这个结果，很多问题，首先agonists应该不是

The SIPSorcery server will be moving to a new host in one week on Sunday the
17th of June 2012 at 0200 PST. The move is to take advantage of some synerg
ies with another SIP service, there will be more information on that further
