never heard back from them. I used the 1" roll and it seemed to be partially
cured as soon as I opened it up - but the package expiration date is 18
months in the future. It isn't pliable, it isn't sticky, it didn't work. It
was very hard to unroll and it wasn't sticky. I used the gloves but didn't

能是因
为里面含有genomic DNA的原因，会比较sticky，虽然跑SDS-PAGE胶的时候sample不是很
sticky，但是跑出来之后做western发现整条lane城smear状，特别是大size的protein
，有人建
议我做一下sonication把genomic DNA震碎，我做了但是貌似不起作用，还有人建议我
treat
with DNase，但是我从来没做过，不知道这个会不会起作用，这里有没有人做过类似的
实验，如果
DNase treatment有作用的话，要加多少量并且在什么条件下treat多长时间呢？
如果这个方法也不行的话，请问还有什么别的办法可以解决smear的问题？

发帖数: 2818 | 10 谢谢你的详细意见。首先要说，我做分子克隆的经验不是很多。其次，基本不会不改变
条件反复重复。
1 当初选用pQE8也是没有办法，因为pQE9我们实验室没有（pQE8和9的区别就是8在酶切
位点之间只有一个碱基，而9要长得多）。最开始的时候没有意识如此近的酶切位点很
难切下来，最起码同时酶切不可能切下来。但是只尝试过一次顺序酶切不行就没有再尝
试用pQE8了，我不记得是先切BamH1还是Hind3，后面尝试插一段片段在2个酶切位点之
间再切。
Nice alternative, shall solve the problem in this case...but I would just do
blunt ligation, nowadays, the fast ligation kits from different companies
such as Roche, takara, don't really care about whether it is sticky or blunt

响一类的，我一个同
学（当时二年级）是物理背景过来读经济的，问他说为什么采用某种sticky price的形
式，有没有去企业调
查过到底是如何定价的，新秀露出很不屑的表情，他的话大概暗示不关心实际上的情况
。实际上采取calvo或者
其他的price setting假设，目的只是为了求得显示解的euler equation或者law of
motion，行内的
人早就习惯了这样的做法，但是一个原来学物理的学生非常合理的“外行”问题就暴露
了这类假设虽然有用，但
是是随意的。
另外一次，听一个IO做得不错的人，讲现实中一个操纵拍卖的团伙的数据，对比拍卖理
论的一些结论，就非常
有意思。
呆在学术机构的人没几个人真的了解现实经济的运行，一些看似puzzling的现
象问学术界的人，他们的第一反应就是guess，很smart的人当然可以列出好些
个理由。说不定每个理由单独列出来，都可以match data，都可以发表。但实
际理由其实就一个，到市场上一问就知道了（当然我是说的简单的现象）。
