车慢，吵，其他不是很清楚。
曾经在D档开动过程刹车座位上笔记本包撞到换挡上，把档从D往前推了一格，相当于突
然换挡.伤gear吧...
我先google下怎么换 transmission fluid, thx! |

去dealer 那做了个oil change +修了一个前灯。
拿回车了发现shift stick 好像比原来高/长出了半个inch.我开始以为自己记错了，可是
我ld也觉得好像是变高了。
车开回dealer, 他们说做oil change+修灯没碰过我的drive gear box，所以不可能。
难到真的是我搞错了？ 有没有大虾知道这可能发生吗？会不会mechanic 把我的车瞎搞
了一把，还换了零件什么的？ |

是265，按理说不至于死这么惨吧，我当时是直接地板油的，发动机转到6K还是看到旁

了，只能在里面很小的空间工作 |

head gasket，rear main seal，timing chain，radiator， water pump等等，现在出
现transmission hard shift，主要在1档换2档，check engine 灯也亮起来了，但code
读出来是P1441 Evaporative Emission Control System Very Small Leak，跟
transmission没关系，因为之前修有warranty，所以会去那家修车铺查，如果是他们的
问题会免费给我修。现在想知道之前修这些leak有可能会影响到transmission吗？想提
前有个数，谢谢! |

我从种子提nuclear extract, 想做EMSA, 用的是DIG-Gel shift kit. 开啥有了
TATABOX （5 repeats) 25bp in length, 试试看蛋白是不是有功能的，结果没加蛋白
的free probe也有带，所以认为我那加了蛋白后产生的带不是真的binding，因为2 带
的位置是一样的。但是有些情况用其他大一点的探针，50bp, 会得到smear. 还有我跑
电泳的胶很小，8x8cm（bio-rad)， 5% native gel. 0.25x TBE buffer, 4C, 80V.
问题：
1）怎么能证明蛋白是用功能的？现在没有binding， 不知是不是蛋白的原因，SDS-
PAGE 检测蛋白还行。抽的几次都是结果差不多，尽管有些带很强且集中在2个区段，但
总体来讲，蛋白的分布大小都有。
2)能不能找个其他probe 做postitive control。 有人可以推见一下啥探针？
3）我是用的nuclear extract, 探针可不可以用200bp 长的。
4)调整些啥才能有好 |

看字面是招NOC，这本身是个低级工种
可看要求貌似还不算太低，真正network engineer肯定不屑去做noc
想做noc的肯定还不符合要求。不知道你们公司咋做的job description
还是个night shift。本人不太看好
尤其不看好老中干这个
