菜单（蜥蜴头），选择电源按钮，里面有log out,
lock screen,power off 等等，一旦选择了lock screen，
这机器就完旦了，症状就是等logout之后，再想login进
入图形desktop, 机器慢得无法使用，几十分钟才能完
成login过程，然后点一下鼠标需要十几分钟才见反映，
非得重装操作系统。这问题我从10.1到13.2都中过招儿，重
装过不下5次。有没有人遇到过类似问题，可知道解决办法？

open an software, when I maximum it, the application always go out of the
screen edge. I have to move it back a little bit then I can see the "Maximum
", the "Minimize" and the "close" button in the top right corn.
It is not very convenient. Does anybody have any idea how I can make the "
Maximum" button exactly fitting the edge of screen. Thanks.

发帖数: 64 | 13 谢谢上面几位的建议. 我也试过用paper towels,但384板上不是每个孔都平均的去掉
medium,所以不行.而且每次靠手来控制力度的话,差异更大. ABI 基本上没什么建议.
如果加一个板子的话,靠人还行,要是加100个板子,根本不可能做到.
做high-throughput screen, 我现在的确是看delta-delta C(t) right, 理论上不担心
每个孔出来到值,但是在284板子上,要用32个positive control 和32个DMSO 做Z
fanctor分析.这样的话,positive control 和DMSO组内的差异必须很小才行.所以只有
组内每个孔细胞数基本一直才行.
还有就是用qPCR 做screen的确耗钱,但现在是花了钱,也不能保证稳定. 真是麻烦阿. |

做Biometrics screening和Health check，如果不做的话就没有Discount，但不影响
plan。
我查了一下，biometrics screening好像是检查遗传信息（不确定，比如癌症和糖尿病
等）。我的担心是一旦做了这样的检查以后，是不是保险公司能够根据情况调整保费或
者拒保？而且这样的信息一旦有了，估计一生都是如此了，是不是很危险？
BTW:好像美国是全球最大的biometrics information provider。 |

别的pooled RNAi screen的时候，在筛选结束要抽基因组DNA。然后PCR扩增hairpin
sequence。再去上array或sequencing，就可以知道哪些hairpin富集了，哪些hairpin
减少了。 我问的是这一步，这时候因为所有的不同基因的hairpin都是混在一起的，所
以不可能得到特定基因的knock down水平 |

很笨的brute force genetic screen 结果是很不确定的，是发散的。你不知道会得到
什么基因。这就是genetic screen 的魅力。你得到了基因，才是开始。
而我说的试错法，是一个一个试你“认为”的可能性，在思维上是收敛的。很可能你认
为的可能性根本就不包括真正的机制。所以随着时间，你会不断补充新的可能性。
我再次说一下，没有绝对的不试错。我也试错。但一个明白人，不会去试所有的可能，
而是会prioritize。
会不会prioritize，会不会分辨，决定你是不是试错法。 |

发帖数: 1553 | 20 哺乳动物细胞里做genome wide crisper screen的就那么几篇吧，都在大牛杂志上呢。
至于Cas9和sgRNA是不是在一个载体上，根据我们的经验，目前最好用的是Zhang Feng
lab的version 2 all in one vector。
有几个实验室载体分开的，必然会有某一个载体的滴度特别低。过这个情况也因为细胞
的不同而异，我们用的是原代细胞，不见得有普遍性。
不过我完全不能理解为什么CRISPR要分成两个载体然后再搞一个inducible system。
从screen的角度来讲，crispr library的数据明显要比shRNA library的数据信噪比高. |

Druker, he will tell you it is rationally designed, muhahaha. Yea, it is a
semi-screening effort and a lot of luck.
Btw, if you are in academia and try to write a HTPS grant, good luck to you.
This also apply to other efforts such as combi-chem and computational etc.
And if you are not in the academia, you better hope your collection is
private and is not in the public domain or element of composition patent is
