一般RIN 7以上就可以了. 你这个只有一条带也有可能是Agilent的Chip有问题. 要是怕
降解就用RNA Later ICE试试.

从微量培养细胞中抽提RNA,没有问题,RIN大于9.2
从微量病人骨髓分选细胞中抽提,少量介于8-9,大多3-8,质量不足以继续测序(for
RNAseq)。
Nanodrop测的浓度都介于20-几百ng/ul,现在浓度不是我最大的问题,RNA降解造成的
质量问题无法解决。
尝试了qiagen的两个kit: Allprep DNA/RNA/Protein 80004;Allprep DNA/RNA 80204
,两个没有大的区别,都是从培养细胞中没问题,抽提病人样品就不行。在犹豫是不是
只能用RNAeasy kit?
问了好几个qiagen的tech support,也分别尝试了他们的方法,均不能提高RNA质量
请教哪位达人有私人珍藏经验可以分享一下吗,多谢了!

大家好,我最近在做RNA seq。样品来自于激光剪切老鼠组织,RIN只有6左右,量也不
多,用了150ng total RNA.True seq V3建的library,qPCR显示library量够多。用
highseq 2500测了,在highthrough output条件下,4个样品在一个lane里,结果每个
样品只得到了10-15M 2X100bp,远低于预计的30M。core facility说他们也不知道原因
。请问大家有什么经验可以分享吗?另外,有没好的测序服务中心推荐呢?

rna定量是用qubit,library是用qpcr,他们说只能这样定量分析了.激光剪切的老鼠冰
冻组织提取Rna,RIN值大家一般能得到多少?有没有好的protocol可以分享

RIN有6已经不错了
fresh frozen LCM关键是切下来立马要进lysis buffer,不过你这个case这些都不是问题
如果core没做错什么的话。。
要不。。truseq。。难道3’没处理好或者adaptor没连好?

这俩区别怎么想?
当然一个interverting,一个non-inverting
两个的gain好像基本差不多,Rout基本差不多
输入Rin呢,可能non-interverting小点??
frequency response大概non-inverting好点因为没gate电容?

可能是因为虽然gain差不多,Rout也差不多,但是Rin的话,还是CS最大,所以好吧??

rin是CS大
Rout也差不多
max vin的话,CS方便,因为S接地,但是如果CG的话,swing上线离gate至少要Vt,所
以不好
ease的话,还是CS方便些。要不G如果接低电压,S就得swing到negative去了
frequency的话,CG好,因为cap小,但是output stage估计多的是low freq了,
不要这个benefit?

for RF applications Rin is not a problem since people can use matching
network or transformers to tune the input impedances so that optimal power
gain can be achieved. Besides sometimes people use the 1/gm property of CG
stage to form a wideband input match to 50Ω.
In low frequency designs when people just connect the input of the second
stage to the output of the first stage it is a problem if the input
impedance of the following stage is too low.

这里的一个重要区别是考虑high frequency, capacitive load
要看impedance,而不只是低频下的resistance.然后用 r*c来近似一个pole.
实际上,也可以假设output resistance 很大,无穷来简化。
你是从vin->vout ,在m3 的gate端的断开的吗?
比如你的boosting amplifier,有Gm, smallest Rout & infinite Rin , ideal opamp
.
从vin, 在m1的输出有 v2, v2=gm1*vin/sc1, 可以画一个current source:gm1*vin //

v2控制boosting amplifier & cascode amplifier
boosting amplifier, 可以画成gm3*v2 //c3, 它有输出电压:v4=gm3*v2/sc3

Enceladus Creates Saturn's E Ring

Explanation: The active moon Enceladus appears to be making Saturn's E ring.
An amazing picture showing the moon at work was taken late last year by the
Saturn-orbiting Cassini spacecraft and is shown above. Enceladus is the
bright point near the center, right near the center of Saturn's E ring.
Streams of ice and water vapor can be seen pouring off Enceladus into the E
