我每次跟老公吵架都会去读关于两性/婚姻/亲密关系的科普书,目前已经读了十本了。
然后有人问,看了十本还吵,是不是证明看这些书没有用?
其实是这样的——读书的用处不在于防止吵架。每个人都是独特的独立个体。人跟人之
间有些分歧是无法“彻底解决”的,是永恒的吵架导火索。很多时候你能做的,是接受
这些问题将永远存在,然后带着这些问题继续生活。
读书的用处在于,读之前我觉得“明明我对他错他为什么不改好气喔”。
读之后我觉得“嗯我自己还是有很多可以改进的地方的。但是仍然是我对他错。他为什
么错还不改呢?因为他蠢啊。”(←此处推荐阅读跟亲密关系无关的卡罗尔·塔夫里斯
的《错不在我》)
一般想到这里,你就能带着平和的心情微笑了。
然后你还会突然理解到佛跟菩萨的慈悲之心从何而来,因为他们眼中的众生都是傻X啊
……

我又called了comcast, 因为上次comcast technician说the dish network cable
inside the current house cannot run comcast, and I need to hire someone else
to rewire the cable line. But I found out the house has the RG6 cable,
which should work for either satellite or cable. The comcast technician who
came last Sunday was not knowledgeable.

duplication致modularity形成的部分,AB也是引用他人的文章。
似乎没有homomeric duplication或interaction rewiring,就不可能得到modularity
。仅仅duplication部分是不够的。

synchronous spiking neuron network, IBM Research在这个方面投了很多钱,甚至把

挖到IBM去。现在这个方向还没有看到有大的突破的迹象。

transcriptional regulation还有phosphorylation是rewiring最快的两个
比蛋白序列的上的变化快
所以我觉得从dna,蛋白序列上找到真正起到作用的差异的几率很小,噪音太多了
同意你说的,RNA-seq, ChIP-seq, 还有proteomics的数据更可信,也更能反应真实情况

部分同意楼主观点。NGS提供更加可靠的数据,对癌症诊断治疗肯定有巨大推动作用的
。但是癌症治疗目前至少有这几个难点:
1. 很多癌症有多个靶点,target一个不光疗效有限,而且复发率偏高;
2. 癌症的亚型太多,而且有stage-specific signatures,这给分析NGS数据提出很大
挑战;
3. 癌症不光是突变在起作用。癌细胞的signaling pathway往往具有rewiring的能力,
而这样的行为很大程度上不受突变的支配;
4. 超大project立项前不光要在科学上证明可行性,还有诸多政策监管上的问题。另外
NGS数据分析不止一种方法,如何在那么多参与人员中达成共识也是个问题。
我对NGS在癌症领域的潜力还是有所保留。当年基因组测序前不也号称攻克癌症不是梦
吗,结果呢?未来20年内全面攻克癌症在我看来是不可能的任务。

2.确实挑战巨大,所以我也提到了需要10多年多国合作。但是反正就是30亿个碱基,就
这么多

1. 抗生素和抗癌药没有很好的可比性。你指的广谱目前除了放疗和一些化疗药外没有
其他选择。即使是一大类癌症,目前市面上的往往也是不能通杀的。比如拿herceptin
去治triple negative breast cancer就不行。不过将来会不会有所谓广谱抗癌药也难
说,比如这篇已经进1期临床了:

2. 加上不同族群的遗传背景,和不同阶段的肿瘤样本,工作量翻几番不是问题吧?
3. 我的观点是rewiring不全受突变支配。肿瘤细胞里的epigenetic modification,

我本人并不是说NGS对cancer研究没用,只是说不会产生革命性的突破~
1)NGS穷举出mutation不会很难,这也是NGS牛逼的地方,但是真正找到driver很难
2)在function研究方面,无论是RNA-Seq还是ChIP-Seq说的都是一个故事:
transcription,一些初步的数据已经证实protein level翻译水平的贡献率大概是2/3,
另外我觉得NGS的定量分析和microarray一样很丑陋
3)我比较赞同进化的观点,因为cancer不能完全归功于mutation~ LS有两位在讨论
reprogrammed signal pathway~ 这个当然有mutation的例子,但是和mutation无关的
例子简直是太多了~ e.g.a lot of weird metabolism pathways will totally rewire
the epigenetic profiling. Some abnormal metabolites can change the pathway.

为什么说找到driving mutation有那么难。我一开始就讲了,现在是做不到,到时候只
要筛的病人,癌组织够多,真正的driving mutation总是会信号越来越强的。因为
driving mutation只要出现了,在它的子代细胞里基本上就总是在那里,类似于我们人
类最后都能用遗传方法追溯到我们唯一的那个母亲,lucy
function 方面的seq我认为不是根本道路。function的东西变化太多
关于你的第三点,首先epimutation也是NGS的强项。另外我不认为epimutation会是致
癌的主流。大部分epimutation根本就不稳定,它在自己那一代细胞里是会造成一些变
化,但你如何指望它都能一代代传下去,造成雪崩一样的本质改变-失去控制的癌变?
起码我没有看到什么好例子

今天收到NIGMS director 的公开信,里面明白着说单个实验室不应该拿到超过三个R01
(除非是clinical research), 并明白指出这个动向的原因是:
1。在钱的总数有限的情况下,一个实验室拿到多的钱就会导致另外一个实验室拿到的
钱变少。
2。大实验室对博士后和研究生培养不利,因为导师的注意力被分散了。
3。从统计上来看,大实验室的成果并不和资金成正比

I guess not much advantage other than ease of deployment and reach of mass
market. How many people are going to completely rewire their house or
apartment with CAT5, and sprinkle plugs evenly through out ? I guess not
many, hence why PLC is gaining popularity.

会疼痛减少。这是stanford 一个group 做出来的。这是个9mil grant, 有图有数据支
持的,不是瞎说的, called rewiring the brain to short circuit pain. The
principle is that perception of pain are highly determined by general health
, heredity, stress, anxiety, fear and depression.
以后大家牙痛肚子痛,就不要浪费钱去看医生,随身多带些照片即可。

It is logic to reason legally blind people may not have input to SCN which
interacts many other regions of the brain. Such loss may disturb the
endocrine system causing mood/emotion problems. But I don't think there is a
consensus over whether legally blind people have circadian rhythm or not.
I don' know the rewiring process after visual loss at the level of brain
cortex. I learned from many TV shows or movies that blind people have
superior hearing ability. LOL

前阵子买了个二手的MB R500.
今天在dealer那里问如果换Xenon车灯多少钱。老头跟我说about 10 grand.
我问怎么会这么贵,他说要换整个head light assembly, rewire,
还要装什么leveling,以免车身不平等时候车灯射到对面人眼睛里
就算这些都加上,10 grand听着好离谱啊。
网上看见一些kit 100多,那些是xenon headlight么?
