scott mills, 81年开始跑ultra, 跑了107个race了....
今年61岁， 这次WS100跑了22小时10分， 他是SD100的race director,
上次training camp他还和我打招呼，可惜那时候不知道他是谁....
反正ultra也不用跑很快， 正好适合老年人....

毕竟0 drop对小腿压力太大...

周的训练计划，从8月中开始准备，可惜训练计划刚过半就半途而废了。。。原因有很
多，起始于训练刚过半的时候打鸡血的跑了个10K，把自己跑伤了，于是歇了一周，从
那以后就开始彻底打酱油，每周跑1－2次。。。比赛前一周索性一次都没跑，心想着5K
反正跑不死，最多慢点，今天没啥压力就去跑了。
最终成绩是27：34，我自己的表测的，官方成绩除去出发的那点delay估计能再少点。
反正一上来就是跟着人群跑，pace 8：30左右，我之前从来没有在这个pace上hold住
3mile，甚至没有过1.5mile的。心想着管他呢，豁出去了！就这么不知不觉跑到中途折
返，开始觉得有些累了，很多人开始超过我，我小心翼翼的保持住速度。2.5mile过后
开始觉得五脏六腑都在翻腾，有点想吐，但眼看终点就快到了，周围很多人在加油，说
什么都要坚持呀～终于终于看到了终点，开始勉强冲刺。倒不是因为我有多少余力能冲
，而是想快点结束。。。跑完后好一阵子才缓过来。。。

barefoot极左，高跟鞋极右。
过渡的话，慢慢来，从右到左，一个台阶一个台阶下，跑到不疼不酸，在下新的台阶。
barefoot和 Minimalist最大差别是cushion，
Minimalist和 Racing Flats差别是heel toe drop。
偶发现偶的脚合适0 drop的minimalist，来一点cushion还是对老腿脚比较仁慈的。 |

发帖数: 7122 | 7 快上吧，真的好！对你这样之前有基础的见效特别快！我第一周就感觉到新陈代谢加快
了，因为没事儿老饿，非常准时的早上10点和下午4点snack时间肚子就开始叫。现在知
道为啥人家一天吃6顿了，因为真的需要。第二周开始早上起来觉得上腹收紧了，早上
空腹净重减到有史以来最低-111lb。第三周明显大腿根粗壮很多（也可以说firm吧，好
听点 ^^），不是自我感觉，ld天天见面的都说壮实了。
我准备6月份纯跑步，多去几个race玩玩，把肚子上脂肪再减减，看看能出来什么效果
。再以后大概方向就是crossfit、tri循环，感觉比去gym拉机器强多了！crossfit都是
group workout，有peer pressure，跑步什么的有race有目标，这样练效果杠杠的。 |

感觉还可以。估计周末的race 破25分应该问题不大。
结果发现跑快了，反而膝盖更舒服一点。可能以前慢跑时跑姿反而不正确。看来以后要
提高速度了。
不过脚后跟还是有点疼，这个问题不好解决阿，希望不要影响周日的race。 |

1）big surf 的半马，据说是最美的 race
2）NYC 全马，这个是必须去的。。
3）雅典马拉松，最正宗的了。。但是最遥远，不行跑个10 k也行
4）keywest 半马，海明威的故居。俺感兴趣
5）上海外滩半马
6）伦敦海德公园的半马
想着凑假期一并跑个半马啥的，挺带劲。平时就在这附近每年跑两个半马。有个念想挺
好。BOSTON那种的不是不想。但是。。。
还有啥跑和玩一起的。据说迪斯尼和RocknRoll的还可以，不过没排上俺个人的list上
。也许以后会变的 |

个前OLYMPIC GOLD MEDALIST来教课并有Videotaping and analysis.我看了一下内容（
见链接），感觉主要是教SKI RACING 的东西比如Giant Slalom and Slalom ski. 而我
的目的是All Terrain Ski 象diamond, double diamond trails和powder ski.不知道

赛的有穿着racer服的hard core racer，也有穿着“便装”的老太太。对于大多数人，
比赛没有所谓的输赢，都是自己跟自己比，非常有助于锻炼提高滑雪水平。喜欢的话还
可以参加race team。

Try to win the race for HU(event 9), and event 17.
But find event 17 has some sick token pro(The Fiend 34) in it. So, I decide to work on HU
race.
Today's HU game is tough, since a lot of high buyin HU regular are there.
My average buyin for HU is only 10 bucks. Those guys' average buyin is at
least 50bucks, some guy's average HU buyin is 400$.
Generally, the field is pretty tough.

多，稀稀拉拉的没多少人，很有些经济萧条的感觉。
第一部Tropic Thunder。 轻松搞笑剧，大牌影星，Meet the Fockers 的Ben Stiller,
Iron man的 Robert Downey Jr.，King Kong的 Jack Black。电影进行了1/5以后出
现一个有些小秃顶的狂暴制片人，脏话连篇，被我看出来是Tom Cruise, 赞自己眼力
了得，主要是他的无理头的莫名其妙的表演方式实在是太独树一帜了。小说原作者，我
误以为是No Country for Old Man里面的Josh Brolin, 后来证实不是。 片子让人从头
笑到尾。紧张工作一个星期之后，还是很需要这种无脑娱乐片去放松一下地。

随机引物可以做正向的也可以做反向的。
但是我有一个方法，是我以前扩增Novel病毒是想到的，而且很有效。
在做反转录时用特殊的随机引物，在随机引物的5'端加一段已知序列例如：5'-
GCTACTAGTCGATCGTnnnnnn-3'。这样你就在所有cDNA的5’端加了一个Tail。
然后做PCR时forward primer用你的5’端Gene specific引物，反向引物用这个tail“5
'-GCTACTAGTCGATCGT-3'”你就肯定能扩到3’端，可以一步一步往3’端扩。
可能ronbell想说的也是这种方法吧？
其实Race最麻烦的还是5’-race。这以前一直用一个酶在cDNA的3’端加A。但不是很有
效，有时可以扩到5’端的序列有时弄很难 |

然后用gene specific forward primer和adaptor specific reverse primer扩增
5'RACE也可以用这个方法做
我试过这个方法的3' RACE，还蛮方便的
因为我做的原核，没有poly-A tail，没有kit可以用
这个方法帮了我大忙

Invitrogen's oligo adapter is about 60 nt，first PCR used adapter 5‘-end ~
30 nt primer and gene specific（GS) primer. Then they use close to adapter 3
'-end nested primer and another nested GS primer.
After the nested PCR, I sometimes still see multiple bands. I have to gel-
purify the PCR products with correct size and sometimes to repeat 2nd PCR to
amplify it before cloning.

1, Check out the anneal temperature. If no band here, but your positive
control showed no problem for RT, then I would suspect that PCR failed in
your RACE. You can try touch-down PCR or use 2-step PCR (68 degree for
annealing and extension.
2, Increase your PCR cycles. If the annealing temperature is not perfect,
increasing cycle numbers will help. I normally used 36-40 cycles for 2-step

求助各位高手。
我用目前的方法做出了2条全长以后，接下来怎么做都做不出来了。
我RACE完的产物每一次都能跑出很亮的1-1.5KB的条带，切胶回收连接到T载体以后测序
。发现序列都是错的，序列两端有制作模板时候加的接头序列，但是中间的片段在基因
组里都找不到，或是一些错的片段。我自己检查了引物，酶，DNTP，水都没有污染，也
不知道为什么会这样，求高手指点. |

说明书上说貌似50min反转录4kb没问题。 我之前做大约10kb的，反转录2个小时，结果
很好。 不过我那个基因表达丰度非常高。如果表达丰度很低的，不知道效果会如何。
另外，建议在预测的TSS后面1-2kb的地方同时设计一个引物做5'RACE， 看看和你从
11kb的位置拉出来的是不是同一个起始位点。 |
