XP13512 是 transported prodrug of gabapentin
由于gabapentin在preclinical有致癌可能，2010年FDA把它拒了。年底重新submit，
PDUFA在4/6/2011。
XP19986是transported prodrug of baclofen.
GERD （gastroesophageal reflux disease)的人群很大，美国估计有60MM, 所以，治
疗GERD的药物市场不小。目前主要

drug delivery的覆盖范围更大点，包括传统的formulation和其他一些现代的技术，比
如prodrug。其实像osmotic pump, needless syringe，drug-coated coronary stent
这种东西也可以叫drug delivery.

ur molecule need to be lipophilic in order to pass the cell membrane(lipid
bilayer), if it is hydrophilic, then u need active transport. but as what u
said, too lipophilic will decrease solubility and cause PK, protein binding
, etc. problems. it is a balance. in some cases i saw solubility gave in
to potency, then people will try to make salt, or prodrugs to get around the
solubility problem |

Acinetobactor to colistin. Also colistin is a prodrug, with a very good
renal function, you might not achieve adequate serum concentration
especially in CNS.
Also, what is Serum creatinine?
Colistin dosing is always a controversial topic. Even in US, it is not seen
commonly.
Honestly, at this point, it is difficult to provide a dose.
