embryo的first branchial arch..整个长度不超过2mm吧。然后把distal part ,
proximal part 分开，体外培养。
楼主需要的agar dish.2% agar加入碳粉让它变黑，然后autoclave之后铺petri dish.
black agar dish 背景解剖容易得多。
finescience的那些工具不错，我们一直都用它。
一定需要人带着做一遍。 |

不到一周，如果除开fish facility自身运转的钱(还不用自己掏)，大概也就花了E3
medium和petri

发帖数: 14844 | 14 我们隔壁实验室作worm的，他们用的显微镜放大倍数低，working distance有好几厘米
，都是直接放petri dish。放大倍数越高，以及对numerical apature要求越高的镜头
，working distance通常越短。而轴向(z)分辨率最重要的一个参数就是NA。所以作z-
sectioning能达到300nm以下解析度的显微镜，比如confocal，objective一般NA都在1.
4以上，而working distance通常不超过0.3mm.
关于折射率不匹配造成的z-distortion，日常生活经验就常见，比如长辈曾经说游泳池
、水塘实际比看着深，不要下去游泳之类。
一般来讲，只要有mismatch就会有distortion，这也是water immersion objective相
比oil immersion的优点之一（盖玻片两边都是水）。这个distortion的修正粗略地讲
就是两边折射率的比值，但是对于大角度的非傍轴光线就不适用了。所以高NA的镜头的

我需要测试我们的peptide对老鼠免疫系统的影响，之前的老鼠实验打完peptide以后老
鼠体内IL6上升。现在老板不想继续用老鼠了，让用mouse whole blood 看看peptide有
没有相同的效果。
我想问这种实验，一般需要多少mouse whole blood? 50ml? 然后买的blood到了以后是
不是一定要当天做实验的，blood一般可以低温保存吗？
还有一个问题是我们认为可能是monocyte会和peptide有一些相互作用，想把monocyte
从whole blood中去掉。常规方法是不是用带抗体的magnetic bead把monocyte拖下来？
我还看到实验说monocyte可以贴塑料，让在petri dish 里培养，让monocyte贴壁，这
个方法可行吗？
新手对血液实验一点概念都没有，先多谢大家建议了。。。 |

growth medium (w/o serum; 1% formaldehyde) in falcon tubes.
If you grow adherent cells in monolayers, it's better to perform the
crosslink in petri dishes.
The basic principle is that: you want to take a snapshot of all kinds of
interactions with the minimum disturbance to the cells. |

这次“王飞跃事件”看来又是中国控制界的一次内斗了。控制界的内斗一直很激烈，主要为以中科院系统所为首的控制理论派和以浙江大学为首的控制应用派的长期斗争，持续了几十年了。系统所一直控制自动化学会（中国自动化学会虽然在自动化所，但在谭铁牛和王飞跃之前只是系统所的一个小弟弟，跟屁虫说不上话），强调理论和文章。浙江大学以孙优贤为首强调应用，很能搞钱，大项目一个接一个，从开始的几十万到现在的动辄几千万甚至上亿。系统所呢只能是几万到几十万或百来万而已。这也就是大家都知道的“系统所有权，浙大有钱”。
