美国东部时间10月27日(北京时间10月28日)消息，位于加拿大魁北克的勃林格-殷格
翰公司但尼尔-拉马利等人在10月26日在线出版的《自然》发表文章，报告研制出了第一
种针对造成肝脏损害的丙型肝炎病毒的药物，并已进行了小型临床试验。他们希望，能使
丙肝病毒的酶功能失活的该种新药，能够最终治愈丙型肝炎，并进而拯救成千上万由丙肝
病毒所致的肝硬化和肝癌病人。
全世界有1亿7千万人感染丙型肝炎病毒，其中约70%的人发展为肝脏疾病。美国每年
有1万多人死于丙型肝炎。目前，干扰素和病毒唑结合疗法是治疗丙肝的最好方法，但只
有不到一半接受该疗法病人的病毒被清除。而且用药治疗后，病人身体虚弱，产生贫血、
发热、脂肪组织损害、抑郁等症。目前，至少有五六家制药公司正在积极研制作用于丙肝
病毒、但不影响人体细胞的药物，以减少药物的副作用。拉马利等则将研制新药的重点，

这个药最早是由 Schering-Plough开发的，这个公司在２００９年被 Merck 买下.
Boceprevir 属于蛋白酶抑制剂，特异性的抑制hepatitis C virus 的一个蛋白酶NS3
protease ，而导致病毒蛋白不能被完全处理而无法组装和成熟.
由于丙种肝炎病毒的寄主特异性和在体内的感染过程的特殊性。该病毒目前没有有效的
体外细胞培养系统，也缺乏可以大规模筛选的动物模型，所以无法采用小分子筛选方式
。该药的开发几乎是纯粹由蛋白结构研究和binding prediction 推导开的，在对NS3
protease进行结晶之后，研究组在一些已知信息的基础上，对蛋白酶的活性基团serine 的侧链进行了抑制物设计
。在得出了最初的蛋白多肽lead 之后进行了进一步的优化，
最终得出了一个最有效的分子结构。

目前各个公司根据HCV 病毒复制途径，开发抑制病毒第二，第三代药物。而且，这些药
物可以和第一代药浑成cocktail。增加治疗效果。

Vertex is a little more aggressive in the pre-market promotions because
Vertex is a company with one product and it is telaprevir. So it has got to
work for them.
