Nitrite is still off the chart.
The water evaporation is too fast, had to add distilled water to the tank
everyday. Added a DIY glass lid to the tank to reduce the water evaporation.
pH: 8.0
ammonia: ~0 ppm
nitrite: 5+ppm
nitrate: ---ppm
calcium: ---ppm
phosphate: ---ppm
temperature: 79F
salinity: 1.024
added more ammonia to rise its concentration to ~1 ppm |

Nitrite started to drop quickly, tank cycling close to the end? So fast (my
fresh water tank took two –three weeks to finish)? Will run for a couple of
more days to confirm.
Added one more dry rock into the tank.
Mixed 5 gallon fresh salt water for water change after the cycling, and the
tank will be ready to host fish (planned a pair of ocellaris clown fish to
start with). The water evaporation was reduced quite a bit with the glass
lid.
pH: 8.0
ammonia: ~0 ppm
nitrite: 0.25-0.5ppm

般这些植物不放在主缸里， 不好看， 主要放在底缸或者背缸里。
高等藻和低等藻（diatom， cyano, etc)的竞争主要也是光线和营养的平衡问题吧。只
要不泛滥影响珊瑚生长， 估计应该没什么。 |

前两天终于迎来了两条小丑鱼: Fancy Ocellaris clownfish, 比一般的Ocellaris颜色
要更红一些， 黑条纹更深更宽。外加两只cerith snail 和三只nassarius snail 用作
clean crew.
刚来的时候有点不适应新环境， 吃的比较少， 今天看着基本上放松了很多， 进食也
比较活跃。录像是他们在吃frozen mysis shrimp. 基本上也没有但心中的打架发生，
两条鱼还比较亲近， 现在晚上host在鱼缸的右下角过夜。
Nassarius snail 状态不是很好， 买的时候就有点犹豫。现在只有一只到处跑， 其他
两只从来不动。在等一两天， 不行就只能扔了。
Cerith snail挺有意思的， 一只到处跑， 另一只把自己埋在沙子里一动不动， 把它
挖出来立刻又钻进去。先不管它了， 看它能藏到什么时候。
准备再进两三只 hermit crab, 然后进一些软体珊瑚， 一只 skunk cleaner shrimp,

bacteria or fungi will most likely also kill your beneficial bacteria in the
tank.
I actually don't recommend use these drugs unless you set up a quarantine
tank for your sick fish. You can actually use Seachem Garlic Guard to soak
your fish food to feed them if they are still eating. It will boost up their
immune system to fight the disease.
On the other hand, your tank is overstocked, and I doubt you have a cycled

我算着票钱值我多少顿午饭钱的时候，压根儿没想到演唱会就1个小时。即使后来
看到韩国的歌单很短，但估计起来加上说几句话和休息时间也得有俩小时。但是真的一
小时就结束了，在全场大合唱Saturday Nights最high的时候，在一个哥们儿手捧鲜花
冲上舞台欲一亲BA芳泽而被十几个保安围追堵截的时候，演出结束了。没唱Animal
Nitrate，没唱The Beautiful Ones，还有好多别的没唱。据说是被邪恶的文化局禁了。
BA的嗓音低了很多，据说唱The Wild Ones的时候假声没唱出来，但我最伤心的是So
Young。这首我听了无数遍的歌，竟然头一两秒都没反应过来是So Young。音调太低了，
而且开场的鼓声没有了，BA的那句激昂高亢的Because也没有了。但是唱到副歌部分的

分化染色的，negative 和positive的都是阳性。
这是protocol：
1.ethanol -20 10min
2.wash 3 times with tap water (这里可以用tap water 么？ 它里面应该有钙吧）
实验员说不能用distilled water, 因为钙会溶解在里面？？
3. 3% Silver Nitrate 30 mins RT 没有特别用光照，就放在实验室里，上面
有日光灯
4. 3 wash tap water
5. 碳酸钙 福尔马林 2mins RT
6. 3 wash tap water

查了文献，多数用的是BD或者Santa Cruz的，但是host是rabbit，我的另一个也是
rabbit，所以希望能避免rabbit。
当然最好不要是mouse的，conjugated的也行。
或者有啥组织学方法可以看nitrate/nitrite level也成。

在的所谓大牛跟他们比起来还是要弱很多，比如王淦昌的那篇Nature好像是诺贝尔奖level的发
现。。。。我觉得这里面应该有从来没发过CNS的。不过，中国未来的希望就靠你了。。。

发帖数: 2405 | 14 you can dissolve in PBS and I think the solubility is up to 10mg/ml in PBS,
if I remember correctly. You can also check the package insert for L-NAME or
the MSDS to find such information.
I make mine at 5mg/ml in PBS and the dose is 30mg/kg body weight.
The half-time of L-NAME varies in literature but the ballpark is around
250min to 300min.
I do i.p. injection and do my experiments after 2 hours. I see an effective
lowering of Nitrate+Nitrite level. Some papers even do it after 1 hour.

你还是测一下nitrite/nitrate比较靠谱。也许你两种注射方式都抑制了nos，只是程度
不一样导致了你的readout差异。
另外，你这个ip的剂量有依据吗？360ul感觉好多阿。。。

发帖数: 1114 | 18 就是这几天一直在帮人做IC 样品，用的标准是DIONEX的7 anion（fluoride, chloride
, nitrite,bromide,nitrate,phosphate,方法上要求的柱子是AS 14A(4*250mm),但是基
于样品持有人坚持相信AS 16A 也可以把7 anion 给分离出来，另外一个原因就是那个
人坚持不肯买AS14,所以就用AS 16A 要求的eluent作了，也就是35mM NaOH,但是run
standard的时候有两个感兴趣的物质还是identify不了，有人建议我换成AS 14A分离方
法上的Na2CO3,NaHNO3作eluent试试，但我总觉得如果两个柱子的成分差别比较大的话
，换流动相会不会对柱子不好。
求建议，谢谢大家！ |

communication board坏了，如果让Dionex technical support派人过来的话要给他们
pay机票还有每小时295刀的service fee,我感觉系里很难一时半会儿决定下来什么时候
派人来。但是目前我急着出数据阿，唉，谁让我在一个小小没钱学校呢.
我想问问大家，我想定性以及定量分析apatite里的正离子Calcium,Sodium, Ammonium,
负离子：phosphate,nitrate and fluoride,我知道IC是最便捷的方法了，但是现在我
能用其他比较好的方法替代吗？我觉得用atomic absorption测金属正离子应该可以吧
，就是得去买灯，ammonium用ion selective electrode应该也可以吧，其他负离子不
知道有什么特别好的办法。请大家帮我出出主意看有没有比较不花钱又比较可靠的检测
手段。

I will work with Ni(NO3)2 powder, make Ni(NO3)2 solution, and some post
experiments with that, and maybe the same work with NiO
I'm green hand with chem, little experience. So except the normal protect
such as glasses, gloves and coat, do I need any other special protect like
masks etc, when working with Ni(NO3)2 ?

model,否则目前的结果很难解释,尤其是只有一次的数据,那个jump不能表示什么.
不同的C对恒定N的影响,就比较容易解释了. |

1） COD的概念是根据electron transfered --折算成氧气的量
2）在转化成COD单位的时候，
所有的chemical凡是消耗氧气的，都是正的（electron donor)
比如污水的COD的值mg/L，就是耗了多少氧气mg/L.
otherwise, electron acceptor, 都是负的;
所以对于氧气，就是-1
反硝化反应，同理，nitrate-->nitrogen,系数也应该是负的

OSHA 40 hr证书，在校期间在学校的Department Lab里工作一年半，做过污水处理如：
RDX降解，DNP降解、Air stripper和UV Vsible处理Amonia Nitrate。
生物方面也做过：微生物再生树脂实验，也是水处理方向的。仪器操作如HPLC、TOC检
测，BOD测定都会独立做，GC-MS学过操作，现在根据实验需要仍在学。在实验室工作期
间很勤劳，实验结果也出了很多，所以导师让我可以在找到工作之前继续为实验室工作。
