phosphoproduct by hexokinase, 2DG6P, inhibits hexokinase activity.
When dealing with metabolic flux, there is nothing that comes clean, that's
why such experiments need to be tightly controlled with multiple inhibitors/
reagents targeting different steps of metabolic pathway. |

glycolytic inhibitor.
There are many other metabolic modulators that you can use, but keep in mind
, they often come with primary and secondary impact of metabolic flux and
you should tightly control your study so that the outcome can be interpreted
properly. |

2. 从你描述的症状来看。应该是不能代谢长链脂肪酸, 没法beta-oxidation
LCHAD=Long-chain 3-hydroxyacyl-coenzyme A dehydrogenase deficiency
VLCAD=Very long-chain acyl-coenzyme A dehydrogenase deficiency
最后导致长链脂肪酸堆积，导致肝损伤，直至肝硬化。
3.避免长链脂肪酸。这类的疾病用Carnitine or MCT (medium chain triglycerides)
oil supplement.跳过长链脂肪酸代谢那一步(请尊医嘱)
4.我想应该是long chain FA氧化通路上的基因的mutation导致的。

If time permits, metabolic abnormalities such as hypokalemia and
hypomagnesemia should be corrected before attempting cardioversion. At a
minimum, hypoxia should be corrected with supplemental oxygen. If a patient
has metabolic acidosis, compensatory hyperventilation after endotracheal
intubation may be indicated prior to cardioversion. Respiratory acidosis
should always be treated prior to the use of sedative drugs. |

acidosis (winters formula 1.5 x 11 + 8) For a bicarbonate of 11 the pCo2
should be around 25 but it is lower (19) so this is not a simple DKA. Second
the patient is unresponsive which could be due to severe metabolic acidosis
so the first treatment should be focused on reversing it as soon as
possible, hence the administration of bicarbonate. |

我家宝宝现在9个月大，前一段时间我们因为孩子需要肝脏移植曾经发帖求帮助。
从4月25日宝宝出院后，我们对他的饮食特别注意，到目前孩子一切稳定。医生们决定
暂缓他的肝移植，不过还在那个list里面，只是不参加排队了。
我们在家唯一做的就是注意孩子饮食，不吃long-chain fat。现在我们越来越怀疑孩子
的病跟他的饮食有关。可是我们能得到的帮助很少，大概因为孩子在西雅图儿童医院
PICU里面的时候他们给他吃了不少long-chian fat，当时孩子吃了后变得很糟糕，我们
任何要求查metabolic的要求都被他们回避了，他们现在就是坚持说肯定不是metabolic
病。
第二就是他的pcp，还在三个月大的时候我们就说孩子脂肪便，特别油腻，是否孩子消

metabolic deisease的DISCOVERY(小分子), CAMBRIDGE主要做CANCER 的DISCOVERY(小
分子), 大分子的DISCOVERY主要在THOUSAND OAK.
我估计主要会裁小分子的R&D. 大分子是AMGEN的强项, 估计不会动. 我个人觉得三番和
SEATTLE最危险. AMGEN一直对CANCER比较重视, 所以CAMBRIDGE不会大动. 三番主要做
metabolic deisease的小分子DISCOVERY,比较悬. 由于PIPELINE 不好, SEATTLE的
development几乎没什么活. 那儿的人都闲的要死. 有一个人在2006参加了4
个MEETING加两个TRAINING.
前两年AMGEN疯狂扩张的时候, 大多数人就觉得迟早会出事, 整个一个几年前PFIZER的
翻版, 只是没想到会来得这么快. 放着好好的大分子不做, 偏要去试小分子.现在他们
唯一的希望就是明年AMG162的NDA了 |

标 题: PhD project 选题求助。

Currently I have two choices for my PhD project.
Two projects (metabolic engineering vs tissue engineering), two different lab.
One is about engineering the microbial metabolic pathway for pharmaceutical
production. The professor graduates from MIT and has been working in this
area for about 7 yrs. Probably he will move to another better university
this coming summer |

发帖数: 117 | 49 系里面有老师在做这个，感觉核心的问题是统计方面没有BIOSTAT的人完全搞不定，
MECHANISM做深入做不过PHARMACOLOGY，感觉就是在不停地做CLINICAL SAMPLE，用用LC
-MS.
不知道有专门做METABOLISM的高手吗？ 现在做METABOLISM 容易找到工作吗？还是会用
ＭＳ走遍天下都不怕。
Transporter 不知道如何？ FDA刚刚出了一个白皮书规范研究，不知道工业界会不会加
大这方面的R&D |
