自交不亲和。一般是先根据同源性设计引物扩部分DNA片段， 然后测序再设计real-
time PCR的引物。拿到引物我会先将Wildtype plant的cDNA用普通PCR仪扩增试一下退
火温度并且跑胶检验40个循环后是否条带单一。确定是单一条带了，会进行大量real-
time实验（Biorad-sybr）。这时悲剧就发生了。有如下一些问题：
1. 同一种cDNA sample的3个技术重复，melting curve不同，有时全是单峰，但不重合
，相差很远；有时3个重复里有单峰也有多峰。
2. 不同cDNA sample之间（WT vs. TG 或者stress treatment at different time
points）melting curve不同，在某个sample里很好全是单峰，在另一sample里就乱了。
3. real-time后跑胶，的确很多带。可是之前用普通PCR扩增，相同program，跑胶是一
条带。

洲也就中国差不多大，相当于中国北京人和上海人melt，本来难度就很低。
接下去少数种族长大了，就不会melt，相反，会变成马赛克，然后会变成南斯拉夫或者
伊拉克，美国内乱肯定是要发生的，黄人少，很可能被屠，大家必须早做准备。我觉得
华人应该集中到一些地区，比如西雅图，联合温哥华，在局部形成多数，这样到时候黄
