you are right in a way, that it would be important to know if this is truly
cancer(though highly likely but without tissue diagnosis it's a still a
question mark), and to know what type,
however, by the scan result, there are mediastinal involvement and thoracs
spine had shown lytic lesions, then it is at least T4, so for non-small cell
it's already stage IIIB, for small cell he could still be limite |

mri not done? ct and mri are definitive diagnostic tests for degenerative
pathologies of the spine.
if it is a compression fracture, one has to find the reason for it:
metastatic tumor? trauma? infectious lytic lesion? osteoporosis? calcium
dysregulation secondary to kidney or other metabolic disease? |

2. What I did is:
Incubate lytic enzyme with your yeast for an hour. Then followed the
BioRad kit miniprep protocol, start from lysis solution (equal to solutionII
in classical protocol). I guess kits from other companies are also work.
At the end, elute your DNA from matrix with 20microliter of TE instead of 100
microliter. One microliter of such extract is sufficient for a bacteria
transformation, even though it's CEN plasmi |

我碰到过类似的情况，不过不是外来的phage contamination，更像是大肠杆菌里自带
的phage进入了lytic cycle。我那次是因为培养箱温度显示出错，实际温度过高造成的
，后来把温度调低了就好了。你检查一下你的培养箱温度看看。 |

发帖数: 6 | 10 我刚开始做金葡菌的致病因子，要用phage从一种strain上载入一个mutant DNA片段导
入另一个strain。用的phage是transducing phage，据提供者说high frequency of
lysogenization。我用plaques提取lytic phage，然后和recipient混合，incubate 20
分钟以后立即稀释、涂板，结果没有一个带resistant marker的colony长出来。
我的实验方法可能会出什么问题？
这种phage是不是主要进行specialized transduction？收集lysogen然后transduce是
不是可行的方法？
