你是做的转基因还是transient 表达？一般我都是先在烟草里面agro-infiltrate试过
之后才做转基因，protoplast也OK，但是麻烦些。
我不同的蛋白连同样的linker（比如说pK7FGW2.0里面的那个GFP linker），表达量相
差10-50被是很经常的事情。有的用35S promoter表达好，有的用inducible （
estrogen）表达好。加不加RNA silencing suppressor差别也很大。有的蛋白老叶子新
叶子都能表达，有的只能在新叶子上表达。
与其在linker上折腾（估计也很难找到万能的），你试试agro-infiltration，用P19做
co-infiltration，这个算是做简单的了。 |
