very simple, just gluten which is the only stuff the ppl w/ celiac dz are
sensitive to.
Many rye,oat wheat grains contains it, and should be strictly avoided by ppl
who have this disease or sensitive to it.
Many shop sale these grains and processed food w/ instruction like "gluten
free stuff"......
someone sensitive to a specific protein, can go to see and concult a
specialist called allery/immuno disease physician who is specific to

If purification depends on traditional precipitation, this gelatin is trash
even it is recombinant human gelatin.
As I know, there are not successful gelatin (recombinant) produced in
industrial for several problems:
1) post-translation modification, which cause allerge and other immuno
problem.
2) Gelatin is a big and complex protein, which means no way or too difficult
to express whole gelatin in pichia or E Coli or oth |

有一些问题想请教有经验的朋友。
我用double staining (uranyl acetate + lead citrate)
一般UA 15min左右，LC 5min，但结果我的sample在低倍下（<1000）很难找到，
高倍的时候背景很重，分辨率不够高。
我2个月前配的UA LC之后，一直锡箔纸包着放4度冰箱。protocol上说可以这样保存好
几个月，但是我觉得最近staining的片子背景有密布的黑点点，应该是lead沉淀。在保
存的时候有什么需要注意的吗?
在配染色液的时候，都是在通风厨里称量吗？好像很不准。但是UA LC不是放射性就是
重金属，不在通风厨里称空气里会有很多粉尘。
在immuno-golden staining的时候，还需要最后double staining吗？我尝试过，但是
背景有时候也有很多黑点，请问如何让背景很干净？
感觉很多经验是protocol上反映不出来的，来这里请教一下做过的朋友。谢谢 |

agonist? Is there known agnoist and you can use? A lot of immuno-system
function on a timing system to determine the final signal, but kinetic these

2. what cell type in pancreas express this gene, epithelial or other?

4. take highly malignant cancer cell lines, overexpress this gene by
transient transfection, cell viability assay, tumorigenecity (in vitro and

give a sh*t about gene/cell therapy because a kid died 10 years ago, they
are getting a lot of funding from big pharmas.
That being said, if you are doing translational stuff like this, it is hard
to get an academic job due to lack of governmental support and the amount of
money necessary to set up the lab. It is also hard to get an industry job
simply because pharma would rather collaborate with labs like Carl's to

还可以直接上CD19 monoclonal Ab，要论杀细胞，我不太明白未什么他们不直接用单抗
治疗呢？
估计大致的回复是：其他方法试验了，都没有好的效果，他们正好撞对了某些东西
还有就是，癌症病人体内有一套机制，遏制免疫反应。有文献报道的，癌人比正常人T
，DC细胞反应弱。理论上，ex vivo culture activating cells before infusion may
not necessary lead to activated cell after infusion.
这些细胞免疫治疗，提高专一性是个大问题。这些个成功报道是有明确的cancer
antigen target。很多时候，做临床试验的，solid tumor Lysis fraction feeding
on immuno cells during ex vivo culture，我个人很是顾忌这种做法，因为这必然用
了正常细胞或广谱的antigen，显然人工制造自身免疫疾病的风险急剧上升。事实上，

wells,and treat them like the normal mounting/floating sections...
Advantage:
1,u could very easily treat the frozen tissues in the well,without running
into the risk of tissue damage,
2, also u could save a little bit more of antibody working solution....add ~
100 ul for each well,
3,also the solution is not easy to evaporate in the humid chamber
4, personally,i think it is better than the frozen mounting slides,which

-chemistry)，主观性很强，每个sample都有亮的区域、暗的区域，很难客观比较荧光
强度。所以想寻找有没有可以用96-well plate来测。
