恩 我是外行随便问问。我有点不理解的是 搞neuron发育的比较直接 因为neuron总
是有分类的(不同的layer 不同的projection map,不同的neurotransmitter等等
) 这样也许可以用transcription code分类然后看他们分别怎么起源和分化的。但
是astrocytes(或者说glia)好像分类不像neuron那么清楚把 这样除了能study这
些细胞整体是哪来的还有啥别的能搞得没

就是说起源都不清楚?我只有个很简单的印象就是glia也从neural progenitor ce
ll来 中间有七七八八的fate determination factor参与。呵呵 我基本对发育问题
的细节知道的是零。。而且好像oligodendrocyte比astrocyte的发育搞得清楚些?

Freda Miller 不算这个领域的吧。glia中包括的很多,发育模式道理上应该是不一样
的。

for Ca2+ P method, make sure the pH~ 7.04; after adding the mixed plamids,
buffer, wait only half hour, then wash out the transfection medium as clean
as possible(you should be able to see only the black aggregates on the cell
surface, but not other places), then add fresh warm culture medium.
You can also try Fugene HD from Roche, which is better and less toxic
compared to lipofectamin and similar products.

问个问题
可不可以在fish里做tissue specific cancer,比如brain tumor,可以在神经系统里特
异KO/overexpress tumor supressor/oncogene来建立cancer model,我知道有人
manipulate fly glia,成功的在fly里引入了glioma模型。fish目前有类似mice的cre-
loxP系统么?

If I am not too out of date, I thought one perfect/classical example is newt
, and even the myofiber can be dedifferentiated and prolfirate again...maybe
reactive glia could be another potential one... Also, in germ lines...many
of these stuff (including EMT etc) are happening naturally, but how
frequently, how easy etc etc is another issue...I think ys ago your boss
together with the other two cows wrote a nice commentary on the natural de-

大概200-300M欧母。
抑制glia的是在培养2天后加的,胶质细胞生长剂。
如何在VOLTAGE CLAMP 下记录NA电流?我们一直做电流钳,封上细胞后就转到电流钳了
。不知道电压钳下如何记录NA电流。

不想让它们分化,就给它们mitogen,如楼上说的fgf2和egf。但是NSC传不了几代就会
自发分化的,要是这时候还给mitogen,glia就会大量地冒出来。

This question was one of my interested topics. To my understanding,
progenitors cells migrate along radial glia within VZ. Once they decide to
differentiate, they migrate away into destined areas. Migration is just part
of the differentiation program. However there are situations that migration
and maturation can be uncoupled.

就算不在乎debris的话,神经元密度实在是太低太低了。周末去不了实验室,周一放假
。周二我再去看看。希望能再多点吧。
神经元是不是有个soma,有processes的就一定是了?有些glia长的和神经元差不多啊。

我好多年没有做neuron培养了,neuron 和glia的大小上是有区别的吧.
neuron有axon and dendrite.
完了完了,第一年修的histology, neuroanatomy都忘掉了.....俺好几年不做动物实验
了,现在开始做人身上的实验了,要是半年后再出不了什么好结果,我就不干research了.
哦,那个"1"肯定是neuron,毕竟俺看了几年的猴脑切片,我以前的几位导师中有一位是做
电极插在活猴脑里的,还要事先训练猴子,猴子可聪明了,还会从我手心里拿东西吃.
你为什么要从化学转到生物上来啊? 化学不是不太难找工作的吗?

就是阻断glia cell的活动,神经系统有两种细胞,一种是神经细胞也就是神经元,另
外一种就是glial cell也就是神经胶质细胞,想找到一种抑制剂,只抑制神经胶质细胞
但不抑制神经细胞

哎,这两天恶补了神经细胞运输问题和睡觉问题,就给大家科普一下吧。
2013年的一篇文章(忘了那个杂志了,好像是nature)说,睡觉的时候老鼠脑子里面细
胞间隙扩大60%,方便脑脊液流动,测得细胞间废物清除速度增加了两倍。所以这个可
能是为什么需要睡觉。
神经细胞的axon确实是需要运输,这个60年代以来就有很多研究了。线粒体都能运,还
是双向的交通。可以运输线粒体,神经递质,细胞膜的蛋白质,细胞内那些线啊纤维啊
的构件都运。运输有快慢两种,就好像火车有快车慢车,慢车开起来也很快,但是要经
常停站。快车就直达,所以比慢车快两个数量级。线粒体和神经递质是快车运的,线啊
纤维啊的构件是慢车运的。
还有人认为axon里面有蛋白质合成,不光是靠细胞核附近合成的。理由包括,不少蛋白
质半衰期短,还没运到就烂掉了。还有些实验证据。又有人认为,神经细胞末端的mRNA
可以来自周围支持的glia细胞。

glial cells 对大多数neuroscientist来说,直接就被忽略不管了。随着技术手段的提
高,比如在glial cells弄chr2等等, glial cells的作用被渐渐的挖掘开来了。。。
想和大家讨论一下,glial cells到底是确实功能上底neuron一等呢?还是不好做,比
如不好patch,杂乱等等。。。。

This is a very interesting area to explore.
The contributions of glial cells (astrocytes, oligodendrocytes and microglia
) are more than what they originally were thought.
I have a particular interest in neuron-glia interactions and their outcomes
(neuroprotection in my case).
Also, gliovascular relationship should not be neglected neither since
astrocytes can form end-feet structures on endothelial cells which are
important for BBB integrity and nutrient supply.

From my personal view, I think anti-oxidant capacity and metabolic coupling
are the two important roles. I don't know which one is the most important.
Also, early reactive gliosis forming scar may inhibit inflammation spread.
But persistent gliosis is bad.
I don't know how dopamine hyperactivity is connected to glia. Would you
please refer me an article?
