说技术含量很低，我老板本科毕业，土法练钢几十年。累死几代TECHNICIAN。但你要参
透FORMULATION这个东西，技术含量相当高，目前的科学水平经常不足以处理这么复杂
的体系。

现在有些专业是研究formulation的, 有的在化工里,有的在药学院里.我其实就是想知
道现在做formulation的人一般学些什么,工作了之后再回顾以前的方向有什么建议. 我
是来问经验的.个例我觉得也肯定有.NONO师姐换了方向,不知道她是否会觉得以前学有
机是失误.
兴趣这个东西我觉得很玄. 我对药物发现是有兴趣的,只是空洞的兴趣. 一旦涉及到细
节问题,特别是以后的工作就是做这一细节而无法掌握全局,我就失去了兴趣.现在就是
这个MedChem. 感觉就是天天合成, 没有一点做药的感觉.可能在这行做下去,辛苦十年,
坐的位置比较高了, 那时候觉得才有兴趣. 但这十年的投资,根据我得到的信息,是很高
的. 投资小于回报, 我觉得我的兴趣就不能再算做兴趣了. 我到底想干什么? 我总感觉
在我没干之前, 可能总也想不到.

印度制药业还是很发达的。有个dr. rebby什么的，据说是世界前50强的制药公司。印
度的API和formulation都做的不错。相对formulation更强大。在美国基本是处于
formulation的统治地位，从学术界到工业界。
在制药行业，印度对中国的优势类似在软件行业。大概就是这样的。
老中在美国制药行业相对在结晶和分析上面更擅长一些。
兄弟，当初老陈要是有长远眼光，不要去搞那个没什么屁用的纳米技物所，instead，
去搞个pharmaceutical technology, or pharmaceutical processing，今天华理还真
能在这方面混口饭吃。 |

我现在在北卡RTP刚开始工作， LD 人在Boston做博后， 刚拿到Cincinnati的一个大的
chemical 公司的职位。现在在考虑两个人是否要我来放弃放弃现在的工作去
cincinnati发展，还是LD turn down offer和我在RTP找机会。 我们现在纠结以后哪里

1. RTP 我在制药领域的 大的CRO工作，在制药领域算是相对稳定。做preclinical的
formulation development的方向。LD可以做制药领域， 但LD在RTP的纯chemical 的工
作不多。现在倒是有在公司做博士后的机会， 但工资不高，老公不是很感兴趣。
2. Cincinnati，LD会在大的化学制剂公司做senior chemist，公司的turnover number

实验室. 看看现在的美国药学院, 几乎没有老板做和工业界formulation直接相关的研
究. 大公司的主流还是片剂, 针剂, 这两年liquid gel比较多, 主要是针对那些low
solubility, low permeability的药.
再说了,就工业界formulation用到的那点技术, 你能完成一个PhD project吗? 所以你
只要大概齐做drug delivery方面的东西, 有点那些physical pharmacy的基础知识, 摸
过,听过,学过那些基础formulation的东西, 找工作的时候你就可以适当发挥了.
如果我是你, 先申请一个drug delivery 方面的好实验室. 不过你国内是生物本科, 申
请drug delivery lab很难, 因为这些实验室几乎只招chemistry background的学生. |

in physical chemistry, biopharmaceutics, material science, and some aspects
of engineering principles.
Otherwise you can enjoy a "easy" life there for just being able to perform

Novartis的Aliskiren，在1995年化合物就搞出来了，但是Novartis做scale up和
process做不好，觉得不可能把那么一个复杂的化合物做到成本足够低到能与别的高血
压药竞争，就放弃了。他们的一个R&D的VP，Alice Huxley，跑出去自己搞了个startup
，Speedel，纠集了一帮人，找了个CMO，设计了新的路线，就能把成本降下去，最后这
个药批下来了，连化合物名字都是Alice用自己的名字去取的。后来Novartis花了将近
一个billion，又把speedel买了下来。这个个例能说明小startup和CMO就比Novartis做
得好吗？
小分子API的development，无论是chemistry,还是process，formulation, 以及后期的

也就是批准4年以后），但是作为新的formulation (象那些XR),新的脂，新的盐，和没
有完全重新做临床用于新的indication的enantiomer，这些non-NCE, 理论上批下来的
第一天就可以file ANDA and PIV。为了抢First-to-File,大家常用的办法就是根据公
布的信息（专利，文章.....)去猜那个药的formulation, PK.....一般都是在PDUFA
Date(brand 批准的时间）之前几个月就把exhibit batch做好，开始stability，甚至
做点pilot biostudy，有的甚至做几个不同的formulation，等RLD可以买到，马上去测
测看dissolution,然后挑出最合适的那个去做BE。过了，马上就file。我见到的最快的
例子大概是在brand批准下来以后40天左右ANDA（因为做BE什么的还是要时间的）就

在一家consulting/agent公司干，公司有几个印度supplier，印度神油的威力我是见
识了，就能吹，“no problem", "certainly"...当面promise的timeline，我加一个月
，都很少有能准时的。做出的东西吗，从质量到严谨性，我都是大大的疑问。
老板想招个新人。找来个老印，开始老板电话里跟他聊了聊，激动得不行，说，你要
赶快跟他聊聊，人很nice。简历转过来，一看，就气不打一处出。上面写着"Ph.D.
expected in 2010",好家伙，看来一年多简历没更新了，虽然我们公司不大，可这种态

看了一下这个版上的ID都是工作了的师兄师姐。最近对自己的职业规划有些迷茫，想向

我现在在美国的州立学校读制药的PhD第二年了。学校制药方面排名还不错，但是我现
在的课题说说实话有些迷茫，老板倾向于做计算模拟方面的。类似于PBPK这种，但是实
验室就我一个PhD是这方向的，本科生物的，对这方面的数理基础不扎实。感觉做起来
特别痛苦。课题进度也很慢，老板也明确表示过不满了。
不过老板还是属于比较nice 的人。因为我编程基础差让我上了好几门cs的课。我问了

比如你说，[1][2][3][4]提出了这个问题，[5][6][7][8]讨论了这个问
题。[8]里面假设x=0，其实x也可以=1, 于是俺解了x=1. 虽然没多大
意义，但是to our best knowledge，这东西没人解过。这种文章审稿
人多半会骂娘。但是反正也没浪费他多少时间，而且要拒也真不容易找
理由。于是就发表了。
要是你说这问题我第一次这样formulate, 十有八九有人会跳出来恶心你。
俺的题目先前有人formulate过，虽然结果很不怎样，没证出啥实质东西。
俺考虑了一个加了限制的问题，然后从另一个角度讨论。敌人上来就说，
你为啥不讨论最general的模型。还说你这方法俺从来没见过，你为啥不用
俺的方法。俺看了review心想，这不正因为您的方法啥也没证出来吗。
总之。好事多磨。只要人家别动邪念，据几次倒也就是个时间问题。

graph only gives you a problem formulation.
You can solve by finding an Eulerian tour. Each node is a 3-digit number eg
100, and two nodes 100 and 001 are adjacent if they can be combined into
1001. Now every node has equal in-degree and out-degree, thus an Eulerien
tour exists and can be found using the standard algorithm. |
