我们是用小鼠胚胎的软骨和骨头染色的protocol。染软骨的话只用Alcian Blue即可。
最后胚胎用BABB处理就会变得透明。BABB危险，而且会腐蚀显微镜，使用时要小心。很
多动物的胚胎都可以用类似的方法染骨头（红色）和软骨（蓝色）：鼠，龟，蛇，鸡，
鱼，蜥蜴，变色龙，蝙蝠。。。而且都很漂亮！

details.
Here is what I used for hepatocyte.
Preparation of the fatty acid stocks
The free fatty acids are prepared by dissolving each individual free fatty
acid in 100% ethanol. Most are readily dissolved in ethanol, though some
require heating to achieve fully solubility.
A final concentration of 100 mM for each of the fatty acids as follows:
31 ul 1 M Palmitic Acid Solid, 1 g / 3.3 ml EtOH

. Four wells of cells are combined into a 1.5 ml EP tube.
3-2 For genomic DNA extraction, 500 μl of cell lysate buffer (50 mM Tris，
100 mM EDTA，0.5% SDS，pH 8) and 10 μl proteinase K (10 mg/ml）are added
into each tube and mixed gently and sufficiently. Bathe the tubes at 55℃

发帖数: 9094 | 11 MeOH and Pd/c is a dangerous combination. Better avoid that combo all
together. Switch the solvent to EtOH would reduce the "fire/combustion"
risk dramatically. When doing filtration to remove the used catalyst, make
sure you do NOT make the catalyst super dry and exposed to air.
I agree EtOAc is a much benign solvent for hydrogenation in this respect.
But the speed of hydrogenation therein is very slow compared to using EtOH.

52 yrs old male, single, lives along, was sent to ER by the girlfriend for
new onset confusion. He was initially hallucinating and become drowsy and
progressively worse over 2-3 days. He just saw his neurologist a day before
and was sent to home, but no details available. He was directly admitted to
ICU.
PMH: ETOH abuse with pancreatitis. But he quits ETOH for 15 years. He has been seeing his neurologist for chronic pancreatits per his girlfriend.

I oppose any sort of habit forming substances except caffeine with
reservation. I hate ETOH which causes too many social problems. Now comes
marijuana, more social problems I predict.
They can set up an age limit but what happens to ETOH will be the same to

用的protocol是４％PFA perfusion fix, overnight post-fix at 4C.
70%EtOH x 2, 95%EtOH x 2, Butanol x 3 (several hours, then overnight, then 3
days)

固定应该够了，难道是脱水的问题？protocol是实验室以前的，现在没有人做这个．

就用100% EtOH 固定加脱水.
我用自己发明的方法通过参照点系列切片法定位细胞. 参照片子可以用任何方法染色,
目标片子不染色, 用100% EtOH 固定加脱水, 然后可以通过参照片上的定位找到目标
片上的细胞. 切下的细胞抽RNA,最高可以达到 9.0 以上的RIN. |

我们用的是palmitic Acid和Stearic Acid两种，分别去treatmacrophage。据说是先用
乙醇溶解，然后再加入BSA，但是我不太明白你这个protocol。
1. 前面那个31uL、2.8uL。。。是什么的体积？
2. 2.8 ul 1 M Palmitoleic Acid Liquid, 1 ml(1g)+2.93ml EtOH
这里面 1 M Palmitoleic Acid是配好后的浓度？ 1 ml(1g)+2.93ml EtOH里面的1 ml(
1g)的1mL和1g分别是啥意思？

MeOH can dissolve nitrocellulose in 10min at 37C.
I never did the soaking before transfer. But tried once after transfer with
MeOH and did not destroy the membrane at least. To be safe, don't soak
nitrocellulose in MeOH.
EtOH is not strong as MeOH. Some type of nitrocellulose is safe in EtOH. |

最近在做纯化m-phenylenediamine，这种东西买来是黑色的固体，纯的应该是白色固体。
查了些文献，只是很简单的说recrystalization from benzene or DCM，可是我试了，
得到的只是黑色的solution。
也有说distillation，然后recrystalization from EtOH的。我distill后得到的是无
色液体，而且和EtOH会混溶诶...
郁闷屎了....

metabolized to acetate by aldehydehydrogenase. Acetate is finally broken to
CO2 and H2O in TCA cycle. Along its various metabolic steps, ETOH
metabolism requires Thiamine as a cofactor and this depletes rapidly.

如果是芝加哥大学医院，当然其它病人比较多。
但是像我们这样被众多nursing home包围的社区医院，icu里大部分就是感染病人，亏
你还是医生，当然你很可能还只是住院医，对美国的真正医疗情况还缺乏直接认识。
哥好歹干了十几年，呆过7家医院，从教学医院到社区医院到psych医院。没吃过猪肉还
没见过猪跑吗?
十几年的veteran怎么也比新警察靠谱一点。
你干长了，呆的医院多了，就知道医院的病人和社区有很大关系。
白人富人区，一堆etoh detox的，黑人区，都是heroin/cocaine overdose的，
nursing home多的地区，感染病人多。
大医院，elective手术病人移植病人多/
至于我们这的cook county医院，那烧伤和枪伤美国最top的。 |

i washed the pellet with 70% EtOH and dry in air 5 min, so it is not too dry.
then dissovle the pellet in water. i think i put enough water. the
concentration will not be higher than 200ug/ml.but a samll damn pellet is
still floating in the water even after 37 degree overnight.
