信号通路对小鼠胚胎干细胞及胚胎早期胚层分化的重要调控作用》（Calcineurin-NFAT
Signaling Critically Regulates Early Lineage Specification in Mouse Embryonic
Stem Cells and Embryos）的论文，该研究筛选到一条在胚胎干细胞（ES）分化过程
中起关键作用的信号通路——calcineurin-NFAT,并对其在ES细胞分化和小鼠胚胎早期

Tumor drug responses 中 ERK1/2 啥的被激活还是挺常见的。比如说手头这个数据，
给药后 5 分钟 ERK pT202 就达到峰值，AKT pT308 也出现峰值，而 AKT pS473 缓步
上升，要在 60 分钟时才达到峰值，同时 AKT pT308 出现第二次也是更高的峰值。而
细胞还是挂掉了。
对了，哪位知道除了 MEK 以外，还有谁能直接磷酸化 ERK1/2 T202/Y204 啊？我们的
数据表明 MEK1 pS217 没有动静，ERK 却发飙了。准备用 U0126 再 validate 一次。 |

但是32-ATP体外磷酸化做了两次，都没信号。
磷酸化实验没问题，因为positive control可以被phos-ERK2磷酸化。
有没有可能ERK不是上游直接Kinase?
不知道那位有好的idea? |

某癌细胞中, 稳转 tet-on shRNA vector targeting gene x (which is a
potential oncogene). 加Dox 3-4天，发现细胞明显停止生长，死细胞增多 (also
there
is induction of PARP cleavage). Akt phosphorylation does not change.
However, these seems obviously increased phosphorylation on ERK1/2 and
ribosomal protein S6. Can't figure how cell death occurs in the
presence of mTORC1 (hyper-) activation.大家说说有哪些可能的survival
pathways, 多谢！ |

growth factors; also, your cells are generally synchronized if there is a
growth arrest with serum starvation. You can skip the starvation, and
usually you'll also see an increase in phospho-AKT, ERK1/2 etc.
You may try 0.2% FBS 0.1% FBS etc. instead of no FBS at all.
Check the literature about the concentrations used for your particular cell
lines. 10-100 nM is a good start. |
