不过我还有些个人看法要补充。我看了新闻，发现美国他们3月份国会（？）讨论的时
候，Howard Univ那个Ron Hira教授说H1B被滥用。有些保守派的反对扩增H1b cap, 觉
得H1b 影响到了American workers, 而反对的声音包括Facebook, MS，Google等大公司
都建议要扩增H1b cap。 我觉得这样讨论来去很难有结果，而且似乎讨论不在一个点上
。大公司要招H1b的人，建议增加名额，他们的理由是“it is foolish to train
foreign students in our higher institutions and give them master and ph.d
degrees, but finally force them to leave the country to create companies

RT-PCR 技术已经推广了有些年头。但从众多已发表的文章中不难看出，很多试验
者的计算多多少少都存在问题。甚至，有些介绍算法的文章都存在明显的错误。因
此，有必要对试验人员讲解计算方法。这样不仅可以尽可能避免计算错误，也能更
好地理解RT-PCR 原理。以下讲解只针对单色RT-PCR，也就是说，在任意一个
well 里，只能用一对primers 检测一个样品。
一、RT-PCR 到底要测什么？
RT-PCR（这里，RT 指 real-time。区别于rt：逆转录）是量化基因表达的一种方
法。如果说western blot 是针对蛋白水平，RT-PCR 就是针对mRNA 水平。
基因表达在蛋白水平上，已经经过了多个数量级的放大。或者说一个mRNA 模板
分子可以翻译出无数蛋白分子。所以，蛋白可以很容易地用抗体探测到。但
mRNA 数目太少，直接用探针检测有困难。所以必须要人为放大扩增才能检测
到。这就是PCR 的目的。

我的问题是：我这个月马上要给NIEHS提交一个R21,我计划6月份的时候把这个R21扩充
一下去申请K22，不知道允不允许？因为这两个proposal是很相似的内容。读了好几遍
NIEHS的K22 instruction,都没有太明白可不可以。
它的instruciton说“Candidates are eligible for a K22 award if they have been
, or currently are, the PD/PI of an NIH R03 or R21 grant or a PHS or non-
Federal award that duplicates the provisions or research goals of an R03 or
R21 grant”。但是又说 “any concurrent RPG application may not duplicate the

对i从0-n-1，不断把a[i]换到a[a[i]-min]去，直到a[i]=a[a[i]-min]。然后再从头检
查一遍前max-min+1个元素是不是满足a[i]=i+min就行。
但他的那个方法是i只从0循环到max-min，并且检测到a[i]=a[a[i]-min]并不停止，而
是把后面的换过来继续做交换。如果观察他的程序，可以看出p只有在出现了duplicate
之后才可能++。p初始是max-min+1，说明我们至少需要max-min+1个不同元素来构成
sequence，每出现一个duplicate，p++。如果p=N就说明我们需要N个元素才可能构成序
列，这个时候如果再碰到duplicate就说明无法取到max-min+1个不同元素了，因为

缘分吧。
题目都是本版的基础题，常见的口水题，自己也练过很多遍。题目都能打出来，但是有
bug。面试之前Collabedit上面练过，但是发现面试时候我编辑哪行哪行重影，模糊，
完全看不清我在写什么，鼠标的位置也是错乱的。只能凭着感觉写，切换到下一行后在
回头看上一行写的是什么。
面试官女烙印，没怎么刁难我，但是我听不懂她她听不懂我。面试时候和她说了我看不
清我写的什么，面完后还截屏让她看到我这里到底是什么情况，希望她能take into
consideration。因为紧张和软件的问题，题目出了bug，HashSet写成Hashset,一个特
殊情况的处理没有把结果加到return set里面。结果一面就挂了， 可能烙印还是在背
后没说什么好话吧。。。

但我提出想看看duplicate怎么处理、设想一下思路，他催促我赶紧写代码，说一会儿
再细说duplicate的情况。
所以我的责任更大。
3那个问题改过来了，笔误。
他之前的文字说法是事先打好的，跟实际说法差别很大，很误导。这个是我比较郁闷的

我的case，f1 opt身份，2011年8月驾照到手，2012年6月OPT过期日过期，2012年3月丢
了驾照，2012年3月去附近DMV，人家直接给我duplicate或者是renewal，貌似是
duplicate，当时没扣分吧，所以没笔试，直接拿到2016年生日当天过期。
最近又丢了驾照，h1b身份。去附近DMV，我没带了护照，h1b的797，但是别人根本不看
，不要，不问。直接说，你要duplicate还是renewal，因为要duplicate要27$，但是新
卡expire 是2016年生日当天过期，renewal只要33$,我一想，还不如办新的，staff告
诉我，新卡5年有效期。但是这次需要我笔试，也许是我有几次交规扣分吧，笔试过，
给我一个临时的。 然后邮寄一个新卡给我。
看了很多人，办驾照，拿什么opt，797，有效期短，贡献一个自己的case。另外，DL
44表格，有个section，for us citizen only, 问你要不要注册投票，你就勾No，我猜

标 题: H1B I797 丢了怎么办？

律师说邮寄给公司了，当时寄出后，他也没通知我们，一直问他他都说他没有收到呢，
让等。但移民局说4月就寄给律师了。6 月份律师突然变成说他4月份就寄给我们了，给
了tracking number ,邮局说deliver了，可是公司没有收到,律师也没有要签名.老板打
电话去移民局，移民局给我们邮寄了一份，但是纸上水印上大大的duplicate.律师声称
移民局只能给邮寄一份原始件，说这个duplicate就可以了。老板也同意。现在我的问
题是如果我回国签证，签证官认这个duplicate吗？我应该怎么办啊？老板和律师都已
经不管了，我打电话去移民局，人家说我是收益人，不能偷漏任何这个case的信息.有
经验的人士帮忙出出主意吧，到底这个duplicate行不行. |

那么，同一个 I-140 可以提交多份 I-485 吗？
移民局说可以，并把这种 I-485 的申请称之为 Duplicate Filing. 而且，Duplicate
Filing 的 I-485 是不用交费的了。
在什么情况下，Duplicate Filing I-485 呢？比如说你的 I-485 在 NSC 搁了一段时
间了，也过了公布的 Processing Dates，于是再重交一份去 TSC，希望能快一点。
除了不用交一千多的申请费，要不要再交一份体检表 I-693？－－不知道。
同样，前面 Second Filing 的时候，体检表 I-693 是新交一份，还是可以指明在另一
份申请中已经交过了，也不清楚。
虽然有 Duplicate Filing 一说，具体是否有可操作性，因为牵扯到跨区递交的问题，
不是很确切。手头连一个这样的例子也没有。 |

法案明说了 USCIS will continue to accept amended or duplicate Form I-140
petitions that are filed with a copy of a labor certification that is
expired at the time the amended or duplicate Form I-140
(3) An amended petition is filed to request a different visa classification
than the visa classification requested in the previously filed petition;
也就是说USCIS接受 amended petition to request EB3 than EB2 requested
previously.
难道不就是解释如何EB2降级EB3么？

Import Photos的时候，一换夏令时，以前import进来的就不认得了，搞得500多张照片
被duplicated，现在还得把他们一一找出来，删掉。恶！！！
I've also had this problem. The underlying problem is how Windows Live
Photo Gallery uses timezones. Repro scenario for the WLPG product team:
Step 1: Take 500 pictures on a camera
Step 2: Insert SD card into your laptop
Step 3: Import all 500 pictures (works perfectly)
Step 4: Take 1 new pictures
