I am rotating in a lab and struggling with the chick embryonic neural tube
injection. I have
already practised for at least 5 times and still could not get it. Now I can
see the neural tube
under the dissection microscope, but I feel really hard to blow and do the
injection. I think the
main problem I have is my failure to dip the micropipette in a correct way.
Or maybe I did not
blow in the correct time point, or maybe the pipette does not have a proper-
sized hole. I feel |

everyone is chasing after optogenetcs, what is your advantage? |

虽然俺不做fish imprinted gene,但是听说很多mammalian的imprinted chromosome的
synteny在鱼里面也都是保守的。当然别人paper作假另当别论。你那个imprinting的机制
,没有哪个能证明吧。。。至少俺是没看见过,等高人答疑。
鱼对于Xenopus的优势无非就是genetics那一点点,能够有很多的突变体,能够做
transgenic fish。injection难度忽略不计,因为每个人都能做,速度不比蛙慢。
microdisection会难很多,但是dissect出来那点点东西不能用来做imprinting吧。另
外就是xenopus个大,产卵却不比鱼多多少,卵黄太多,不太好做生化。
当然,鱼比起老鼠的那一点点优势也就在于能做发育早期一个很小的window,xenopus
在我看来是更没啥可取的,连那一点点window都因为不好做fluorescence消失殆尽。 |

you can use 91460-11 x1 to cut open skull from the top carefully without
damage the brain.Then take the whole brain off.
I have no idea why you need thin wax board.
We put 2% agarose in a small tissue culture dish.You can put the brain with
PBS in the dish for farther micro-dissection with tool 11255-20 x2
91500-09 x1.

embryo的first branchial arch..整个长度不超过2mm吧。然后把distal part ,
proximal part 分开,体外培养。
楼主需要的agar dish.2% agar加入碳粉让它变黑,然后autoclave之后铺petri dish.
black agar dish 背景解剖容易得多。
finescience的那些工具不错,我们一直都用它。
一定需要人带着做一遍。 |

Actually, I found the mineral zones in the middle attach firmly after
decalcification, while chondrocytes and hypertrophic chondrocytes are easier
to detach after in situ incubation compared to no decalcification tissues.
