发帖数: 154 | 4 我用那东西来测定Cl, SO4 和NO3。三周前，发现background conductivity 上升到了1
.4us 左右，以前在相同的Eluent浓度（15mM)和相同的current (50mA)下，那个电导应
该在0.7左右。而且在1.4us的背景电导下，测定的sensitivity就比原来低了很多，测
出的数据明显不准。当时打电话给dionex support center咨询，一个女人说这是由于
那个supressor需要再生，或者就是那个supressor太老了。我们这个supressor到明年6
月份expired。我当时就用0.2N的硫酸进行了再生，发现再生后背景电导下降到了0.6左
右，但是在样品测定中出现了retention time的漂移，原来Cl的retetion time是在6分
钟多一点，现在移到了7分钟多一点，硫酸和硝酸的retention time也移动了1分钟左右
。另外，我们的程序设定在10分钟到18分钟之间Eluent的浓度从15mM上升到45mM，所以
baseline应该相应的在10分钟的时候就开始缓慢上升，但现在虽然浓度是从10分钟的时 |

发帖数: 1114 | 10 就是这几天一直在帮人做IC 样品，用的标准是DIONEX的7 anion（fluoride, chloride
, nitrite,bromide,nitrate,phosphate,方法上要求的柱子是AS 14A(4*250mm),但是基
于样品持有人坚持相信AS 16A 也可以把7 anion 给分离出来，另外一个原因就是那个
人坚持不肯买AS14,所以就用AS 16A 要求的eluent作了，也就是35mM NaOH,但是run
standard的时候有两个感兴趣的物质还是identify不了，有人建议我换成AS 14A分离方
法上的Na2CO3,NaHNO3作eluent试试，但我总觉得如果两个柱子的成分差别比较大的话
，换流动相会不会对柱子不好。

用的，但是总是出现pump over pressure 这样的问题，之前觉得是柱子和guard
column的问题，就从dionex重新购置了新的柱子guard column，但是装上之后还是有这
个问题，看了trouble shooting,似乎也都不是原因！真诚希望得到各位高人的指点。

来。我已经给dionex的technician service 打了很多次电话，他们都说啥样的
suppressor 配啥样的tubing就行，除了flow rate不一样，其他的好像没什么区别，但
是老板还是觉得肯定有advantage和disadvantage，想找出我们到底应该用2mm的
suppressor还是4mm的？

2mm suppressor压力会太大，2mm柱子配4mm suppressor也会back pressure不够影响分
离。

communication board坏了，如果让Dionex technical support派人过来的话要给他们
pay机票还有每小时295刀的service fee,我感觉系里很难一时半会儿决定下来什么时候
派人来。但是目前我急着出数据阿，唉，谁让我在一个小小没钱学校呢.
我想问问大家，我想定性以及定量分析apatite里的正离子Calcium,Sodium, Ammonium,
负离子：phosphate,nitrate and fluoride,我知道IC是最便捷的方法了，但是现在我
能用其他比较好的方法替代吗？我觉得用atomic absorption测金属正离子应该可以吧
，就是得去买灯，ammonium用ion selective electrode应该也可以吧，其他负离子不
知道有什么特别好的办法。请大家帮我出出主意看有没有比较不花钱又比较可靠的检测
手段。

很远（大概两分钟，短柱子），但是后来用ICS-3000试的时候，这三个离子的保留时间
相差很小（大概0.1分钟）实在想不通为什么，我用了一个DIONEX AS12A柱子在ICS-
3000上试了一下，钠和钙的保留时间相差很近，好像说明系统没有什么问题，用的同一
根柱子，中间可能相差了5个月，有人能帮我解释一下吗？ |

helpful to demonstrate what your chromatogram should look like.
Sodium and calsium supposed be separated with much better resolution, unless
your mobile phase was too concentrated.
Describe your conditions may help trouble-shooting.
Don't know what are you trying to do, if you are trying to use the ICS3K to
evaluate your packed column, I would use a known good column, run a 6

能比 linearity range很小的 所以建议你把sample都dilute到类似的水平 如果从0.
04 mg/l开始 最高的standard也就到4 mg/l吧 不然肯定不准。。。 |

发帖数: 154 | 30 实验室里用一台DIONEX的离子色谱仪来测定水中的氨，在状态好的时候它甚至能测出DI
中的氨，结果前段时间在连续运行某一批样时，DI的波峰逐渐消失，然后标准样的波峰
越来越小，在接下来的2批样中，发现标准样的波峰还在继续下降，一开始以为是柱子
的问题，然后换了个新柱子上去，结果这个问题还是存在。请问，这可能是什么原因造
成的吗？头痛死了，机器不合作，然后后面还有一堆样要赶着测，有个年度报告还等着
这些数据。

高，而不是象现在卡在1左右，而且也没有任何漏水现象。
那个仪器我们今年已经买过1个阳离子的supressor，阳离子的column和guard column，
阴离子的eluent，花了我老板4000大洋。我老板现在一听到要换那个部件就心疼。再这
么攒下去都快可以买个新的了。
你说的注射部件是指autosampler那里的取样器还是其他？
对了，supressor再生是怎么一个原理？为什么要用硫酸而不是盐酸？ |

发帖数: 1168 | 36 注射部分就是手动注射那个套筒和里面的金属以及塑料圆块, 用来和injection loop
tube一起精密控制注射到column
里面的实际样品体积, 那个磨损时间长了就会导致同一样品数据重现性极差, 换了后就
好了
再生就是清洗那些污染物残留 (金属离子什么的, ) 用什么酸肯定是公司测试过的最优
选择吧, 我倒没想过为什么 |

conditions.
How big is that peptide mssing on column? sometime big peptide would just
be too hydrophobic, change to C8, C4 column for separation would help.
And I think he was using a Dionex Ultimate 3000 with trpaping column,
whether the peptide can be efficiently trapped is also a question.
