2.是否已知其调节发育？
3.是否揭示了其调节cyclin水平的机制？就你的描述蛋白水平只是揭示了相互作用和表
达，没有完整清楚的机制。
每个问题都可能会影响所能投杂志的一个档次。 |

是的。其实分子通道研究比较多了，但是我们研究的非通道功能至今没有报道。
是的。基因敲除小鼠模型已经建立了。但是机制未知。我们描述可一种可能的机制。
没有涉及cyclin，但是涉及到下游check point分子，which和我们研究的通道蛋白有相
互作用和表达上的关系。

你的数据可以这么解释
d1太多，导致不成熟的细胞提前进入s期，通不过s期检验点。
可能你首先需要做个同步化看看是不是加速进s期，rb磷酸化如何
然后看看dsb如何，s期检验点查什么东西。 |

细胞在G2的时候，由于各种因素造成DNA损坏（过氧化，辐射，端粒太短等），或者癌
症基因例如RAS表达（此时没DNA损坏，这个效率比不上前面各种因素），都引起P53表
达，P53表达引起P21表达，P21的水平高到一定程度就会把cyclin B1关进细胞核不出来
，加上其他P21影响的其他途径（cdk1（次要）, RB（比前者重要）， 等）导致细胞跳
过M，直接进入G1。这样的细胞是4倍体，如果继续细胞周期则会先8倍体然后分裂；因
为多个centrosome或者其他原因会导致细胞染色体数目变化，癌变危险性大，所以有必
要制止这样的细胞继续细胞周期。这里P16就上场了，它通过RB等途径把细胞限制在G1
，不再进入S。这细胞就老化了。老化的细胞有分泌变化自我标记，等着免疫系统来清
除。人老了免疫系统变差，老化细胞越积越多（这一步暂缺因果关系），各种组织和器
官老化就都来了。
还有疑问的几步：
1. 看胚胎发育时候的老化细胞是不是4倍体（估计不是，因为没有P16表达）。虽然依

看了一批文章，总结这个故事最make sense。修改下不准确的地方。加reference。

因表达导致SSB或者DSB等），引起P53表达，P53表达引起P21表达，P21的水平高到一定
程度就会把cyclin B1关进细胞核不出来【Krenning 2014】，加上其他P21影响的其他
途径（cdk1（次要）, RB（比前者重要）， 等）导致细胞跳过M，直接进入G1【Ye
2013, Johmura 2014】或者某些别的致癌基因表达，虽然没有DNA损坏，但是只要引起
DDR，也可以导致细胞跳过M，直接进入G1。或者，强制引起P53表达，或者强制P21表达
，或者什么别的原因导致细胞跳过M，直接进入G1

我问这些问题是因为之前和一哥们儿聊过一些。他做AP后其中一个方向研究细胞周期中
的3D genome变化。需要找一个实验模型，要至少几个million的细胞，而且是高度同步
的。虽然使用物理和化学方法可以是培养的细胞同步，但是我总觉得会不会有一些
artifact。
我倒是有个很好的系统，某种动物胚胎发育过程中，前3个细胞周期天然高度同步，而
且从一个个体就可以拿到几个million个胚胎，关于cyclin的诺奖甚至都是在该物种里
最早发现的，可见很适合研究细胞周期。这种动物的进化地位其实比果蝇线虫更接近人
。但是问题在于这个系统不是主流模式生物，genome虽然在06年就发表了，后来也有不
断改进，但是很不完善。而且很多抗体估计都没有人在这个物种里开发和尝试过。考虑
到种种风险，我的同学决定还是上保险靠谱的动物，先做老鼠和人的细胞吧，就没有跟
进了。
各位评价一下这样的系统是否可以用来研究3D genome在细胞周期中的变化？ |

里面的应考技巧也很管用。还有，我觉得Pharmacology最重要的是记住药名的后缀，就
