发帖数: 387 | 2 Biotin-ABCDEFG（Biotin在N末端偶联的一个15短肽）。
做的是这个短肽的浓度梯度包被，从50微克每毫升开始，4倍梯度稀释，共10个梯度，
明胶封闭，Streptavidin-HRP二抗显色。结果如附件所示
浓度稀释的越低，反应反而越高，Streptavidin-HRP二抗对不包被抗原的孔是阴性的。
但是值得一提的是，我们的阳性对照，Biotin-IgG，来包被的时候，也发现最高的反应
浓度是在包被的一个中间位置，而不是包被浓度最高或者最低的浓度。
请大家帮忙分析下。 |

可是问题是我label的蛋白一直load不上去，load到的信号非常低。。。。 我用了
maleimide-PEG4-biotin 去label cysteine，结果一直不好。信号就是很低的时候就达
到平衡了，很奇怪。 之后我换成NHS-LS-biotin，这个大家用的比较多，结果load的信
号更低。。 我都有BSA去test labeling，都信号很低。。
我现在的问题是，蛋白肯定是label了，30%左右，我用的1 biotin： 1 protein的
ratio做的labeling，因为不想over label multiple sites on one protein. 都做了
两个月了，快绝望了。。。什么条件都试了，Octet red的信号就是不行！
另外，有人说BSA 不好label，想问问别人有用其他蛋白label的吗，我想做个positive
control， 说明是蛋白的问题，还是label的方法的问题

发帖数: 1630 | 4 多谢。终于找到明白人了。我dialysis没有那么多 。我用500x 1h Twice. 这个可能真
的不够。可能有free biotin在 。 下次我一定试试你的dialysis的方法。我octet 信
号才3-4 nm 有的时候更低。就是很低就饱和了。确实说明binding site没了。
另外我是label 4°C 过夜。这样就成了三天时间label一次了。。。
另外我用的是maleimide-PEG2-BIOTIN 50mM HEPES buffer. 标记SH group
另外我也有NHS -LC-BIOTIN... 本来想用BSA 去label做个control 结果bsa也load不
上去
听了你说的。我感觉真的要用更多的dialysis

You could add avi tag to your protein DNA sequence and express it in the
mammalian cells(293T).
Octec company have the biotin tips, and you can purify the protein and use
the biotin tips directly. dont put biotin into your cells. It would be
copurified with your avitag protein and interfere the following binding
assay. |

Wiki了一下，说Biotin食物里面不富含，pantothenic acid肉类食物什么里面都有不会
缺。

发帖数: 47 | 22 我想把没有biotin的DNA（400bp)从有biotin的DNA（100bp)中分离出来。 之前做small
-scale的时候用的是NEB的streptavidin magnetic bead，效果非常好，而且很方便。
但是现在做large-scale, 感觉太贵了。之后试了pierce的streptavidin cartridge 和
resin, 都没有NEB的分得干净，而且还有20-30%的损失。我不确定是我操作不得法，还
是pierce确实没有NEB好用。
烦请前辈给推荐个好用的产品或者指点一下方法。

发帖数: 1604 | 23 我用的是pH 7.4的20mM Tris Acetate buffer，还含了140mM的NaCl和5mM KCl, 1mM
CaCl2, 1mM MgCl2
这个recipe应该是从这篇paper来的：Bock et al. Nature 1992. (355)564-6. 这篇
paper不是用biotin-streptavidin interaction，但是我们就用这个条件做好像没啥太
大问题，至少bind得貌似挺好。
你有没有做个positive control看看是不是你的nanoparticle或者labeled oligo本身
的问题啊？biotin-avidin的binding应该是不太挑条件的，要不然就不会应用那么广了
。 |

猜想，可以用一个反义短DNA链做模板，用polymerase加多个biotin-T在3’端。

干净，要不immobilization会很差，哪怕一点点，这个是小分子，比protein

筛选。具体而言，对于一定浓度的antibody，用1倍，2倍，。。。20倍biotin试剂来修
饰，然后用western blot从中找出最多biotin的活性antibody。
所以你的antibody失活，不是Pierce的错。 |
