富的大侠们对此的看法。
实验大致上是用喷雾剂喷出一些virus stock suspension，接着用gelatin filter收集
这些带有病毒的小颗粒，然后把gelatin filter溶解在beef extract溶液中，最后用
infectivity assay测病毒的感染性，以及用qPCR测总共病毒的量。
照理qPCR测的是病毒DNA/RNA，而在雾化和空气传播过程中病毒又会失掉一些
infectivity，所以qPCR结果应该要比infectivity assay得到的值（TCID50或PFU）要
高，但是尝试了几种动物病毒和噬菌体之后，qPCR给出的结果都要比infectivity
assay要低（有的甚至低大于一个数量级）。
qPCR的standard curve就是用已知浓度的virus stock系列稀释之后做的。考虑过
gelatin filter和beef extract对viral nucleic acid extraction的影响，也做了几

，说一个东西对proliferation没影响，但对cell death有影响（忘了他们是用flow还

非直接的， 甚至可能都不在你primer的范围内。crosslink大大降低抗体的识别 （即
便多抗也有影响， 说没影响的， 做个WB试试就知道了）， 所以对抗体要求还特别高
（估计要用全长蛋白做的多抗才好一些， 可是现在很多都是用重组肽段做多抗的）。
唯一的好处， 这个算是in vivo， 要是shift assay的话肯定是 in vitro了。 in
vivo的话 应该还可以用luciferase assay + 1.过表达/抑制 TF, 2. 突变/deletion 结合序列 来做， 我倒觉得这个更可
信。 |

因为是第一次用这个药物，在准备flow样品之前，显微镜下观察细胞很happy，没有任
何要死或已经死了的迹象。对于这个cell line，PEannexinV/7-AAD Assay的voltage及
compensation都是原来调好的，并且这次实验的positive control和negative control
也很好。可一上flow惊讶地发现经药物A处理过的细胞齐刷刷地飞进了UR double
positve象限。
我之所以怀疑这样的实验结果是因为，首先细胞镜下看没什么异常；FSC/SSC也很正常
，不像是一堆死亡细胞
这是一个很常用的assay，请问版上前辈，该怎么理解这样的结果，多谢 |

因为以前没有做过，所以向各位有经验的大侠请教，希望不吝赐教。
问题1:
luciferase assay和GFP相比有什么优势啊？如果直接在promoter后面接一个GFP，直接
FACS看GFP的表达不也可以看基因表达的变化，而且还不用任何特别的试剂盒。
问题2:
看了Thermo fisher网站上的信息，有不同版本的luciferase。比如firefly
luciferase只能表达在细胞内，需要细胞lysate，然后加入luciferin，ATP等才能测发
光。 而一些其它的版本，比如Gaussia Luciferase，可以直接分泌到media，这样不用
提lysate，可以直接测发光，方便多了，为什么还有很多实验都用的firefly的呢？

最近比较倒霉，需要测量 adenylate kinase 的 activity。换句话说，
需要测 ATP 或者 ADP 或者 AMP 浓度隋时间的变化。一般来说用
NADH coupled enzyme system (PK/LDH) 就可以了。因为ATP
的浓度很高，而PK本身在高浓度ATP下自己就有很高的activity，所以
这个寻常方法不能用。哪位大牛能指点一下？
