核算可以amplify,但是如果蛋白可以给你足够量的样本,也不需要amplify。
我觉得MS还是有些问题不能很好的解决。比如DNA/RNA fragmentation可以通过机械作
用/离子等作用等来尽可能的“随机”打断DNA/RNA来保证coverage的均一性。但是MS就
必须用酶来在特定位点来消化。
而且现在很多DNA/RNA测序为了解决扩增带来的bias,好多测序方法都是amplification
-free的。
不过我又仔细想了一下你这个amplification还可以理解为测序过程中的互补配对边合
成边测序,这样的话这个amplification是决定性的,在illumina、PacBio等测序方法
中。
但是oxford nanopore, ion torrent在测序过程不需要利用这个互补配对合成的过程,
当然后者的library prep中可能还是要用到。所以oxford Nanopore是完全和
amplification无关的。
所以关键还是ATCG四种信号相对简单易测,加上各种修饰也还好。蛋白几十种氨基酸加

cycle是两倍。如果你的efficiency不是2,比如是1.85,那么3个cycle就是2^3和1.85^
3的差别,也就是说实际6.33倍的差别被当成了8倍的差别。这是你的internal control
。如果用这个去adjust你的target的差别,加上target本身的efficiency如果也不是
100%,这个真正的差别就会非常大。
第二个问题是amplification是S型曲线,就是说,在开始阶段,或者在东西很少的不在
中间一段linear范围内的时候不能用delta delta Ct。
我的意思是,cell number不是可信与否的关键,关键是amplification efficiency。
在你怀疑你的结果是否可信的时候,建议用standard curve instead of delta delta
Ct来计算。只要你的sample的Ct落在你的standard curve里面,而你的standard curve
的correlation ef |

I had some real-time PCR run with TaqMan probe. A false positive result was
showed for an actual negative sample. When I checked its amplification curve
(green line in figure below), I found it's different from other samples.
The signal intensity never goes up as a real amplification curve (of even a

efficiency? You input cDNA is total cDNA, you don't need to adjust every
time, but you do need to check your primer linearity and amplification
efficiency. If you didn't, please tell that to the reviewers, lol. Btw, I
have seen primers that are good in taqman that are completely useless in
SYBR rxn.
Btw, show me you actually validate your amplification efficiency and
linearity before we keep talking.

如何自制消音伴奏 by 东海
原唱MP3找不到的情况比较少,歌曲伴奏找不到却是常有的事情,尤其是一些通俗老歌
,要么没有伴奏,要么是以前VCD格式下粗糙的单声道伴奏,或者LD盘粗重的主旋律捣
乱。怎么办?自己试试消音。具体的消音原理这里不提了,大家GOOGLE一下就可以找到
,我只说我自己是如何制作消音伴奏的。前提,PC上安装了CE2.1(2.0)加上至少
Ultrafunk fx插件。BBE和WAVES我在消音时没有用到。
消音伴奏我常用的方法分两种,一种是CE自己带的,在Effects->Amplitude->Channel
Mixer,选vocal cut,通过调整New Left Channel和New Right Channel的百分比来控制
原声的保留程度。原唱制作得比较精良的,左右声道伴奏混音又不多的,可以用100%消
,否则伴奏损失惨重。那时需要消到大约90%处(具体要听出来的效果)。另一种是

哦,酱紫,那真的只有一个copy啊。。。试试楼上说的,nest三轮吧。别的真的没啥好
办法了。
另外,可以中间加个probe,用qPCR做个test,看看amplification efficiency。
你看,DNA的平均分子量是330g/mol,如果100bp的product,那么平均分子量是660g/
mol。agarose gel加EtBr,100ng的DNA应该可以detect了吧?那么100ng大概合10e11个
copy的DNA double strand分子。
咱们如果assume PCR的效率是100%,那么就是那么一个copy要amplify到10e11大概需要
37个cycles。。。
如果PCR效率比较差,比如只有50%,那么需要62个cycles,所以啊,你看,最好先测一
下你的amplification efficiency。 |

质)上扩增成序列一样的的足够多数量的分子。 这个过程产生的 error 很小,比如 <
0.1%, 可以忽略不计。说这个过程可以纠正 error, 是因为在后续 sequencing 的时候
是读取的 amplification 之后整体产生的信号,所以即使这个集合里面某些少量的分
子产生错误的信号, 也淹没在 noise 里,不影响集合读出的正确序列.
相比较之下,single molecule 的方法只读取一个原始分子的信息,比如 nanopore,
读错了就错了,没有这么一个纠正的机制,所以就只能靠大量的 coverage 来弥补。
根据 nanopore 的网站,他们用某种 dsDNA binding enzyme 在 nanopore 的端口来
unzip double strand, 所以最后通过 pore 读取的是 single stranded
trinucleotide。一个分子被测序之后不会回来反复被测。所以要靠测序的 coverage

intutively 看不是万能的,至少我没这本事。 画画small signal有些时候还是很有用
