I would agree that X = Br would be much better than X = Cl.
Another suggestion: I would use NHBoc instead of -NPhth to protect the N in
your alkyne. Phthalimide is an imide, which is not very stable under strong
basic or nucleophilic conditions. Lots of these tough Pd-coupling reactions,
like your case, require high temperature and strong bases. NPhth may
decompose before the alkyne couples. Considering your other pyridazine is

然后和LYSINE反应,不过一般会有很多LYSINE. 最后,如果你想特异地连在某个位置,而
蛋白中CYSTEINE也有多个的话,你可以用SCHULTZ的办法在你想的位置引入一个KETONE,
然后和HYDROXYLAMINE或HYDRAZINE DERIVATIVES反应.当然你也可以引入一个AZIDE或

a good point, but I beg to differ. since silylation involves Si-H and Si-CH
=CH2, Si-O contribution should not be pivotal? if it is, then it is mainly
due to steric hindrance after si-o bonding with the alkyne alcohol?
hope you can make more comments. any reference on Si-O quick bonding? thx! |
