大家最看好哪一类？有没有哪一类现在没有blockbuster，但将来有望出现blockbuster
的药物?
我看好bcl-2的抑制剂，即诱导细胞凋亡的抗癌药，现在Abott有一个化合物ABT-737，
已经进入临床试验阶段。希望该类药物能成为将来的blockbuster!

用靶点的新结构类型的化合物，来取代目前的statin类，一旦实现突破，成为
blockbuster肯定没问题。
目前研究较热的PPARalpha 和PPARgama的抑制剂好像没有足够信心。 |

generate blockbuster if including anti hyperglycemia. If you look at the
top 200 brand and 200 generic drug sales, and top 300 prescription, you will
get a clue.
1st class is cardiovascular, 2nd is CNS, so far. Even a BPH one is easier to
crack 1B than an oncology one.

此外，Abbott还有一个ABT-751,应该是作用于tubulin的化合物，也进入了临床研究阶
段。

Kosan Biosciences Inc在开发hsp90的抑制剂17-AAG。此外还有一些其他的生技公司如
Serene,Synta等都在做。
这一领域，KU有个教授工作做得不错，此外，Sloan-kettering也有人在做，做得不错
。Danieshelfsky好像也做。

Anyway, for cardiovascular disease, many effective medicines have already
been developed. For anticancer, anti-HIV drugs, the medical need is still
unmet.

I do not like the biphenyl, sulfonyl group and nitro-benzene substructure in
ABT-737. It might be such groups cause some side toxicities or bad
bioavailability profile and Abbott finally decided to advance another
compound into clinical trials. |

而且必须和现有的一些抗癌药合用。这是个big problem. |

From what you said, I think the failure of this compound is related with
poor bioavailability for the oral administed preparations. I guess it does
not meet "Rule of 5".
