electrospun nanoyarn fabricated with two methods for the application of
urethroplasty

ABSTRACT:
Urethral stricture is a common disease caused by over-expression of
extracellular matrix from fibroblast. However, the fibroblasts is a main
type of cells for the repair of urethra.Here, we compared two nanoyarn
scaffolds with the characteristics for improving fibroblasts infiltration
but inhibiting the over-expression of extracellular matrix. The Collagen/
Poly(L-lactide-co-caprolactone) (P(LLA-CL)) nanoyarn scaffolds were prepared
by dynamic liquid electrospinning and conjugated electrospinning
respectively. Meanwhile, co-axial electrospinning technique was combined
with the nanoyarn fabrication process to produce nanoyarn scaffolds loading
Wnt signaling pathway inhibitor. The scaffolds morphology were observed by
scanning electron microscopy (SEM) and mechanical properties of the scaffold
were examined. Fibroblasts were cultured on the different nanoyarn
scaffolds. Cell morphology, infiltration and proliferation on the scaffolds
were investigated by SEM, hematoxylin-eosin (H&E) staining and methylthiazol
tetrazolium (MTT) assay, respectively. The release rate of different drug
delivery scaffolds were also tested with high-performance liquid
chromatography (HPLC). The results indicated that cells showed and
considerable infiltration and organized morphology along the nanoyarns
prepared by dynamic liquid electrospinning (DLY). It was also observed that
the DLY significantly facilitated cell proliferation. Moreover, the process
of dynamic liquid electrospinning did not affect the efficiency of drug
release. In conclusion, the D-DLY could faciliate the infiltration of the
fibroblasts but inhibit the collagen production. The D-DLY could be a
promissing scaffold for the treatment of urethra stricture.
