枯草杆菌感受态细胞的制备及转化
1) 将菌种接种在LB平板上37 ℃培养过夜。
2) 用接种环接一环菌苔于5mL GM I 溶液，在30 ℃慢摇床（125 r/min）振荡培养
过夜。
3) 次日取2 mL转接到18 mL GM I 中，37 ℃快摇床（250 r/min）培养3.5 h。
4) 再取10 mL上一步骤的培养液转接到90 mL GM II 中，37 ℃慢摇床培养90 min后
5,000 g，10 min离心收集菌体。
5) 用10 mL原培养液上清液轻轻悬浮菌体，悬浮后的菌体即为感受态细胞可以直接
用来转化。
6) 感受态细胞的保存：加30%的灭菌甘油至终浓度为10%，混匀后分装到离心管中（
0.5 mL/tube），随即放到-70 ℃保存。
7) 转化时取出离心管，放在45 ℃水浴中溶化，然后在0.5 mL菌液中加入适量DNA（
～1 ug/ml）。
8) 于37 ℃振荡（200 r/min）保温30 min后涂布相应抗性平板，再在37 ℃培养过
夜，次日检查和验证转化子。
枯草杆菌普通化学感受态细胞制备用试剂
[1] 10×最低盐溶液：K2HPO4 70 g，KH2PO4 30 g，（NH4）2SO4 10 g，（
Na3C6H5O7•2H2O）5 g，MgSO4•7H2O 1 g，在蒸馏水中依次溶解，加水至
500 mL。
[2] 氨基酸溶液（根据不同菌株的基因缺陷型准备）：10 mg/mL，贮于棕色瓶内，
113℃灭菌30 min，用黑纸包裹。
[3] GM I溶液：1×最低盐溶液95.6 mL，20%葡萄糖2.5 mL，5%水解酪蛋白0.4 mL，
10%酵母汁1 mL，10 mg/mL氨基酸溶液0.5 mL（50 μg/mL）。
[4] GM I溶液：1×最低盐溶液96.98 mL，20%葡萄糖2.5 mL，5%水解酪蛋白0.08 mL
，10%酵母汁0.04 mL，1 M MgCl2 0.25 mL（2.5 mM），1 M CaCl2 0.05 mL（0.5 mM）
，10 mg/mL氨基酸溶液0.1 mL（5 μg/mL）。
