样的WCL？
如果是单一蛋白，我不知道怎么解释。如果是WCL，我觉得
是你的二抗坏了。二抗坏了的时候与样品中的蛋白有很多非特
异性的结合。封闭剂对这个有影响，但不同的封闭剂影响
不一样。所以你看到不同大小的带，但是都是非特异性结合，
我觉得肯定不是你要看到的蛋白。
建议换换二抗，用新鲜一点的。 |

1xloading buffer), 可是那些稀释过的样品lane就变得很宽, 把没有那些稀释过的样
品lane挤得很窄,而且变成smear了, 是不是因为总蛋白的量差的多, 才会这样啊. 那为
什么是蛋白少的跑道挤蛋白多的跑道呢? 而不是反过来呢? 是不是因为盐离子浓度的关
系啊? wcl我是用PBS悬了细胞, 然后加等体积的2xloading buffer的.
问题在哪呢? 该怎么做才行? 谢谢. |

1. in vitro expression: myc-tagged protein-A IP Flag-tagged protein-B 这是普
通IP过程。
2，如果分开表达的WHOLE CELL LYSIS做IP，那说明这2个蛋白相互作用更加直接(对上
下游信号要求不严格)，很可能是一级结构结合。
反之，就是需要正确折叠或者磷酸化修饰等二级三级结构。
3. 一起转染表达的话，根据表达量来决定IP需要的WCL ug数.最常见IP：400-500ug
WCL加1ug抗体。 |
