验中，这个结合位点只结合G这一个蛋白；ChIP也证明G可以结合到F的promoter;如果把
这个结合位点突变掉，基因F promoter的活性在Luciferase assay中几乎全部失去 （
低于2%）；但是现在把G 用siRNA knockdown后，虽然G的mRNA下降了40%（还没有检测
蛋白质）但是基因F的mRNA只下降了20~30%，是因为siRNA knockdown的太少么？有没有
别的可能？有经验的XDJM能不能给指点一下？谢谢！ |

发帖数: 35261 | 5 1.你克隆promoter的目的是啥？如果是看有没有转录因子调控的话，需要看看ChIP-Seq
等数据，确定enhancer包含在里面了。单纯minimal promoter只要几百bp就够了。
2.克隆模版有两种。一个是自己提纯genomic DNA。1500bp基本上不需要kit，裂解然后
土法提纯一下就p得出。运气好的话直接拿裂解液p都能成功。更好的方式是用BAC做
template。B以前基因组测序的时候别人就把genomic DNA切成小片段放到载体里做成了
library。去blast一下就能得到包含你目标序列的BAC clone。买回来摇菌提质粒，然
后用质粒做模版p，效果比用genomic DNA高很多。
3. 设计引物。一般是取upstream promoter sequence到第一个exon。如果第一个exon
有TSS，引物设计在TSS之前。
4. vector。不知道你做reporter construct的目的是什么，不过luciferase好像多用

正经的promoter克隆，如果做功能的话，你说的几个方法一个都不对，或者说全是野路子
你随便拉一个做真正promoter的人出来，第一告诉你是用bac或者其他库，酶切。
虽然你也提到了bac但真的不是你那样用的，只有实在没有办法了，才走PCR的路子。
