1. 如果你的蛋白有aggregation，加一些Triton100(0.1，0.5or1%). 听很多人说，这
很有用。
2. 用4-12% TBE gel （Invitrogen）。如果自己配，用4%。或者3%，下层加三分之一
15%防止free probe run out。
3. 如果还不行，用垂直配胶系统（Bio-rad配PAGE gel用的， 1.5mM）配Agrose胶（1%
？ 2%？记不清了）。里面可以加Triton & glycerol。
我的蛋白80kD， oligomerization 后500 or 1000kD （FPLC测定）。6%TBE gel完全在
well上，4-12%能进一点点，3%可完全进入但很难看。最近我们lab新来的Postdoc用了
Agrose胶，效果不错。我让他加了一些Triton & glycerol，效果更好了，binding
affinity 都增加了几倍。但具体浓度，我得问问他。 |

东西的用途和以前的钙离子成像的用途几乎一样重要。
细胞内的氧化/还原状态一向是一个非常重要的话题，尤其是整个生物领域往生理倾斜
的过程中，如何对氧化/还原状进行定量评估一直是个重要问题。现有的几个工具（比
方说那几个荧光蛋白）都有背景过高而且不准确的问题。你们做的probe如果能解决这
个问题的话会有大用处。
不过，这里有几个问题你们可能需要解决：
1。 对于细胞有无毒性？尤其是长期培养
2。 反应速度如何？
3。 可逆性够不够高？（这个会决定你们的定量线性程度）
至于在动物内部，这个目前不需要太着急。只要能够对肿瘤这种小局部的组织能用就行。 |

，要做但也不急。我个人比较关心tissue culture上能不能用。毕竟很多probe发明出
来，isolated cell culture看着还行，上到组织水平就不好用了，于是在生理水平的
应用就有很多限制。不过总的来讲我觉得您这个发明还是很有前途的，好好优化推广一

. Some cells if you treated them with high glutamate, it will block
cysteine uptakes and deplete its GSH level. Your probe has a Micheal's
acceptor, so I you will have toxicity around 10s microM, but it is probably
okay to do short term live cell imagining.
I wounder if you treated with certain compounds, would you be able to
determine specific GSH pool depletion because there is a cyto- and a mito-
pool of GSH.

发帖数: 183 | 45 最近在试RNAscope，ACDbio,用的是RNAscope® 2.5 HD Detection Reagent-RED
老鼠的皮肤石蜡切片。用了Kit里的Positive Probe, Negative Probe, 我的target
probe, 还有以前同事的一个已知work的probe也做阳性对照。试了4次了，
用同样的片子试几种probe，现在的情况是：
positive probe 看到强的signal，dots,就算work了。
target probe 也看到些红色signal,算work。
但negative probe 也出现一些紫红，虽然明显比target probe 弱，理论上Negative只
应该有Hematoxylin的蓝色。
用同事的probe的片子，很强的信号在皮肤的真皮位置（是期待的结果），但问题是我
的片子在皮肤的表皮，也看到一些信号，这个不是期待的，同事以前用这个probe做老
鼠的片子，表皮只有Hematoxylin的蓝色。
感觉Negative和同事probe的结果，出现的不想要的信号，可能是同样的原因引起的，

经典物理的开山之作“自然哲学之数学原理”发表于1687年。书中，牛顿第一个讲到的
物理实验是水桶实验。
牛顿说，用一根长的软吊绳提一桶水，把吊绳拧成麻花状。如果你握住吊绳，不让麻花
状的绳子松开，桶及桶中的水是相对是静止的，水面是平的。突然放开手，麻花开始放
松，吊绳旋转，水桶也随着吊绳旋动。最初，桶中的水并不转动，只有桶在旋转，桶和
桶中的水有相对转动。慢慢地，水被桶带动，也开始转动。最后，水和桶一样转动。这
时，水和桶之间又是相对静止的，不转动的。但水面却呈凹状，中心低，桶边高。牛顿
