的猫妹，她是我的第二只猫。她三个月的时候我从PETSMART领养来的，是为了给猫儿子
作伴，我一手抱大的。小时候和我亲，不抱就不睡。长大后就不理人了，她会来蹭我，
偶尔跟我撒娇，来找我睡觉，但不许我碰她，不许我主动拉她来一起睡，梳毛时候身体僵
硬，态度也很勉强。但可以一起玩。后来我发现她不是不喜欢我碰她，是她胆子很小，
性格过于腼腆，加上她哥哥经常欺负她欺负的很厉害，所以我碰她的时候她都会吓一跳。
我的猫儿子，以前是人家放养的，就是说每天都随意跑出去和邻居猫一起玩的
那种。后来人家不要了（其实他就是我从这个版上领养的），我带到我家，我住
的是封闭式公寓，不可能让他出去，出去肯定就得走丢了。于是他就开始郁闷。
大家都知道猫是爱清洁的动物，经常用舌头刷洗自己。我的猫儿子因为郁闷后来
都不清理皮毛了，身上的毛经常是乱糟糟的，黯然失色的。每天除了睡觉就是耷
拉着脑袋站在门口等着人放他出去玩，我当时很替他痛苦，都想让他出去算了。
可是出去的结局就是走丢和被车轧死，我没办法接受这个。于是就领养了前面那

I remember reading somewhere, that they sell a pellet, which includes all
premixed supplement, bone and organ, all you add is muscle meat. maybe that
was in my dreams.
if just supplement, it is easy to buy taurine, salmon oil, vitA B and D ba.
for me the easiest for now is 50% can 50% raw, that way dun have to worry if
100% right, the cans have plenty supplements.
i hate handling both liver and heart. |

remain the same, then there is nothing to worry about.
If you really really want to make sure, there are things you need
to check:
1. did they change the fluorescent lamp/detector in the realPCR machine?
2. Did you use the same DNA dye in the PCR reaction? (if the dye is loaded
in the premix kit, then it is not consistent between batches and cannot be
used to do direct comparison)

You are totally right....The relative Ct seems remain same and the dye
actually is added together with the premix. The reason why I worry this so
much is that the Ct value of my target gene is so late (>35) even though I'
ve already increase the concentration of cDNA so much. |

buffer也
就那几样东西，统统换新的，有钱的直接把acrylamide也扔了
如果跑的marker是premix的，上样buffer可以排除了

实验室一年就用20管感受态，那当然是买划算
如果像我们这种三天两头要买感受态，一个月大概要花$1000在这上面的呢？
如果你要一天200个平板，定得更make sense
（我说称重是说LB培养液哈，和那种premix的LB Broth Powder比）
我说的是有时候随手就能省的事情，可以多做做
假如有一天谁要海归，没那么多经费的时候，是不是也会采用这样的策略呢 |

发帖数: 28562 | 17 没啥特别的， 就是把各种东西，包括buffer, dntp, primer,water, sybrgreen, DMSO
, taq
polymerase 都先混合好作为母液，然后分装到管子或者孔里去。最后再把template 加
进每一个管子里，然后离心甩一下。
其实就是把公司的premix 自己装一下。 不知道这个是不是你想问的？ |

还是不行的话，可以考虑换enzyme或者buffer

我买的这个,蛮好用的.
以我的经验,10ML的最好用.一次只要洗4桶就好了,要是严重就多洗几下.
PS 我是一个月就丢一根,所以我从来不消毒.
